响应面法优化微波-超声波辅助提取茶皂素工艺研究

         

摘要

The microwave and ultrasonic assisted extraction of tea saponin from defatted oil-tea seed cake was studied in this paper.Under the condition of ultrasonic power 80 W, radiation for 50 min, the effects of ethanol concentration, liq-uid-solid ratio, microwave power and extracting time on tea saponin extraction yield were studied. On the basis of single factor experiment, the optimized extraction process of tea saponin through the response surface method, on tea saponin yield response value, was studied. Tea saponin was purified by reversed phase column chromatography and was characterized with IR, UV spectra. Experimental results show that the optimized extraction process was: the concentration of ethanol was 77%, the sol-id-liquid ratio was 1∶20 (g∶mL), microwave power was 714W, extracting time was 336s. At the optimal condition, tea saponin extraction yield was(8.1±0.3)% and its purity was up to 65.2%. The microwave and ultrasonic assisted extraction showed synergy in this experiment and tea saponin of highly purity was obtained by reversed phase column chromatography.%以油茶籽粕为原料,乙醇水溶液为浸提液,通过微波-超声波辅助提取茶皂素,在超声波功率80W,超声波作用时间50min的条件下,考察乙醇浓度、料液比、微波功率和浸提时间对茶皂素提取率的影响,在单因素试验基础上,以茶皂素浸提得率为响应值通过响应面法优化茶皂素的浸提提工艺。以反相柱层析法纯化提取的茶皂素,对纯化的茶皂素以红外、紫外光谱进行表征。结果表明,超声波功率80W,超声波作用时间50min的条件下,茶皂素的提取工艺为:乙醇浓度77%,料液比1∶20(g:mL),微波功率714W,浸取时间336s,在优化条件下,茶皂素浸提得率为(8.1±0.3)%,其纯度为65.2%,微波-超声波辅助在浸提中表现出协同作用。反相柱层析法纯化提取的茶皂素,可得到高纯度茶皂素。
