皮肤来源间充质干细胞对内皮祖细胞募集效应的体外观察

摘要

目的 细胞的募集趋化与归巢被认为是间充质干细胞(MSCs)修复重建受损组织的重要机制,本研究拟通过体外实验观察皮肤来源间充质干细胞对在血管化中发挥重要作用的内皮祖细胞(EPCs)的募集效应.方法 分离与培养胎鼠皮肤MSCs和大鼠EPCs,利用免疫组织化学法,分别检测二者的SDF-1与CXCR4表达;并利用体外迁移实验,观察MSCs是否影响EPCs的迁移.结果利用免疫组织化学检测,MSCs和EPCs可以分别表达SDF-1与CXCR4;利用双室培养系统发现,MSCs可明显促进EPCs的迁移,且阻断SDF-1表达后,这种促进效应得到抑制(P<0.05).结论 体外实验证实了MSCs可以促进EPCs的募集趋化.%Objective Recruitment is considered an important mechanism for mesenchymat stem cetts (MSCs) to repair and construct damaged tissues. This study was to observe if mesenchymat stem cetts ptay important rotes in chemotaxis of endothetiat progenitor cetts ( EPCs) in vitro. Methods The skin MSCs of fetat rat and rat EPCs were isotated and cuttured. The expressions of SDF-1 and CXCR4 were detected by immunohistochemistry. Migration experiments were used to observe whether MSCs affect the migration of EPCs. Resutts According to the resutts of immunohistochemistry, MSCs and EPCs can express SDF-1 and CXCR4 respectivety. MSCs can promote the migration of EPCs and if the SDF-1 expression was btocked, this promoting effect woutd be suppressed (P<0. 05). Conctusion It is confirmed that MSCs can promote chemotaxis of EPCs in vitro.
