卵泡刺激素对上皮性卵巢癌细胞株的增殖及β-黏连蛋白表达的影响

         

摘要

Objective To investigate whether FSH can induce the proliferation of Ovarian Cancer Cell Lines and the effects of different levels of FSH on expression ofβ - catenin in these ovarian cancer cell lines. Methods Cultured SKOV3 and HO8910 were treated by FSH with different concentration and time. The absorben-cy of the cells were detected by methyl thiazolyl tetrazolium( MTT ). The effects of β - catenin expression were detect with immunofluorescence by taking FSH of best concentration and time as experimental group and equivalent cell culture medium as control group. Results FSH could induce the proliferation effects in ovarian cancer cell line SKOV3 and HO8910. When the concentration of stimulation was 40mIu · mL-1 for 48h, the difference of the two cells was statistically significant( P <0. 05 ). Positive β - catenin expression was found in SKOV3 and HO8910 cell lines. After being stimulated by FSH, the β - catenin expression increased signifi-cantly( P <0. 05 )in SKOV3 and HO8910. Conclusion FSH can induce the proliferation effects and the expression of β - catenin in ovarian cancer cell line SKOV3 and HO8910.%目的 研究卵泡刺激素(FSH)对卵巢癌细胞株的增殖作用,观察FSH刺激后各卵巢癌细胞株中β-黏连蛋白(β-catenin)的表达情况.方法 不同浓度 FSH 分别作用SKOV3 细胞和HO8910 细胞不同时间后,MTT 比色法测定各组吸光度值.以FSH最佳刺激浓度和时间为实验组,等量细胞培养液为对照组,用免疫荧光方法研究FSH对各细胞株中β-catenin表达的影响.结果 FSH可以促进上皮性卵巢癌细胞株SKOV3 细胞和HO8910 细胞的增殖,在刺激浓度为40mIU·mL-1,作用时间为48h时,两株细胞均增殖明显(P<0.05).β-catenin在两株细胞中均表达,且经FSH刺激后两株细胞中的β-catenin均表达增强(P<0.05).结论 FSH可促进上皮性卵巢癌细胞株SKOV3 细胞和HO8910 细胞的增殖,并引起其β-catenin表达的增强.
