CXCR3B在结肠癌组织中的表达及对结肠癌LOVO细胞株迁移能力的影响

         

摘要

Objective To explore the change of the expression levels of chemokine CXCR3B in paracancer tissue and colorectal cancer tissues and its effect on migration ability in colon cancer cell lines LOVO. MethodsDifferent colorectal cancer and paracancer tissue samples were collected and immunohistochemistry (IHC)method was to detect the overexpression levels of CXCR3B. qRT-PCR were used to detect the expression of CXCR3B mRNA in NCM460 and LOVO cell lines. LOVO colon cancer cell lines was overexpressed the CXCR3B. Wound healing and transwell migration assays were used to detect the migration of LOVO cell lines. ResultsImmunohistochemistry revealed CXCR3B positive expression in paracancer was significantly higher than that in colorectal cancer tissues group(P<0.001). The expression of CXCR3B mRNA in NCM460 cells was higher than that in LOVO cells(P<0.01). Wound healing and transwell migration assays showed that overexpression in CXCR3B group was significantly lower than that in NC group(P<0.01).ConclusionCXCR3B expression is low in human colorectal cancer and CXCR3B overexpression can inhibit cell migration of LOVO.%目的 探讨趋化因子受体CXCR3B(CXC型趋化因子受体3变体B)在结肠癌组织中的表达及对结肠癌LOVO细胞株迁移能力的影响.方法 常规收取结直肠癌及癌旁组织标本,用免疫组织化学(IHC)染色方法检测CXCR3B的表达;实时荧光定量PCR(qRT-PCR)检测正常肠上皮细胞株NCM460及结肠癌细胞株LOVO中的CXCR3B mRNA表达,过表达CXCR3B-RFP慢病毒感染LOVO细胞株,划痕实验及transwell实验检测LOVO细胞株的迁移变化.结果 免疫组织化学结果显示在结直肠癌旁组织中CXCR3B的表达显著高于癌组织(P<0.001);NCM460细胞中CXCR3B mRNA表达高于LOVO细胞表达(P<0.01).划痕实验及transwell迁移实验显示过表达CXCR3B组(LOVO-CXCR3B)细胞迁移较阴性对照组(LOVO-NC)低(P<0.01).结论 人结直肠癌组织中CXCR3B低表达,过表达CXCR3B可抑制结肠癌细胞株LOVO的迁移.
