高效液相色谱-串联质谱法同时测定生物样品中酪氨酸及其代谢产物

     

摘要

A simple,selective,rapid and reliable method for the simultaneous determination of 3,4-di-hydroxyphenylalanine (DOPA),tyrosine (Tyr),4-hydroxyphenyllactic acid (PHPLA)and 4-hydroxyphen-ethylamine (TA)in human serum by high-performance liquid chromatography (HPLC)coupled with tan-dem mass spectrometry (MS/MS)was proposed for the first time.Under optimized conditions,all com-pounds were resolved within 6 min.The calibration curves were indicative of good linearity (correlation co-efficients >0.998 9),and the limits of detections and quantifications for Tyr and its metabolites ranged from 0.000 5 to 0.20 mg·L-1 and 0.001 to 0.40 mg·L-1 ,respectively.Recoveries of Tyr and its metabo-lites in breast cancer patient were in the range of 85.2%~114.0%,with RSDs of 0.7%~8.5%.The pres-ent method was successfully applied to the determination of DOPA,Tyr,PHPLA and TA in sera from breast cancer patients.%建立了高效液相色谱-串联质谱法测定乳腺癌患者血清中多巴、酪氨酸、对羟苯基乳酸和酪胺的分析方法。在已优化的质谱条件下,待测物在6 min内完成分离,且效果良好。结果表明,4种物质的检出限在0.0005~0.20 mg·L-1之间;定量限在0.001~0.40 mg·L-1之间;线性相关系数在0.9989~0.9997之间,线性关系较好。血清中4种待测物在3个不同加标水平下的平均回收率在85.2%~114.0%之间,相对标准偏差在0.7%~8.5%之间。以上结果表明,该方法准确可靠,可用于实际样品的测定。
