DNMT1在卵巢癌组织中的表达及其对卵巢癌细胞增殖和迁移的影响

摘要

Objective:To explore the expression of DNA methyltransferase 1 (DNMT1)in ovarian cancer tissue and its effects on the proliferation and migration of ovarian cancer cells.Methods:The levels of DNMT1 in cancer tis-sues of 31 patients and normal tissues of 15 patients were detected with immunohistochemical analysis.Overexpression of DNMT1 by transfected pcDNA3/Myc-DNMT1 plasmid and knockdown of DNMT1 by transfected DNMT1 siRNA. Proliferation of SKOV3 cells was detected by CCK8 method. The expression of DNMT1 was measured by Western blot.The effect of DNMT1 on the migration of ovarian cancer cells was detected by Transwell assay.Results:The ex-pressions of DNMT1 in 31 patients of ovarian cancer(83.9%,26/31)were up - regulated than in normal tissues (20.0%,3/15).The expression of DNMT1 in cancer tissues of patients with clinical stageⅢ-Ⅳ,pathological grade G3,and distant metastasis was up-regulated significantly(P＜0.05).The growth and migration of SKOV3 cells was significantly promoted by overexpression of DNMT1,and the growth and migration of SKOV3 cells was inhibited by downexpression of DNMT1.Conclusion:The expression of DNMT1 is significantly higher in ovarian cancer tissues than in normal ovarian tissues.DNMT1 can promote the proliferation and migration of ovarian cancer cells.%目的:探讨DNA甲基转移酶1(DNMT1)在卵巢癌组织中的表达及对卵巢癌细胞增殖和迁移能力的影响.方法:采用免疫组化方法检测15例正常卵巢组织及31例卵巢癌组织中DNMT1的表达水平;利用DN-MT1的过表达质粒及DNMT1 siRNA瞬时转染SKOV3细胞系,Western blot检测DNMT1的蛋白表达水平, CCK8法检测各组细胞的增殖情况,Transwell小室实验检测DNMT1对卵巢癌细胞迁移能力的影响.结果:31例卵巢癌组织中 DNMT1阳性率(83.9%,26/31)显著高于15例正常卵巢组织(20.0%,3/15).临床分期Ⅲ-Ⅳ期、病理分级G3、远处转移者DNMT1阳性率明显升高(P＜0.05).过表达DNMT1后,SKOV3细胞增殖能力及迁移能力均增强;敲低DNMT1后,SKOV3细胞生长及迁移能力受抑.结论:DNMTl在卵巢癌组织中的表达明显高于正常卵巢组织,其具有促进卵巢癌细胞增殖和远处转移的作用.
