LRP16与ART-27的相互作用的鉴定

摘要

目的:鉴定LRP16与ART-27的相互作用,推测LRP16基因在肿瘤发生、发展中涉及到的信号途径.方法:酵母双杂交系统筛选出ART-27可与LRP16相互作用,采用酵母双杂交回复验证、免疫共沉淀及GST pull-down等体内、体外方法对二者之间的相互作用进行验证.结果:LRP16与ART-27在体内、体外均可形成复合物.结论:ART-27是LRP16的特异性结合蛋白,二者的相互作用为进一步研究LRP16在胚胎发育及肿瘤发生、发展中的功能及信号传导途径奠定了基础.%Objective : To identify the interaction between human LRP16 and ART - 27 , and to speculate the signal pathway of LRP16 on the development and progression of cancer. Methods : The interaction of ART - 27 with LRP16 was found by yeast two - hybrid screening. And this interaction was tested by co - immunoprecipitation analysis in vi vo and GST pull - down assay in vitro. Results : LRP16 could bind ART - 27 in vivo and in vitro. Conclusion: ART - 27 might be a binding partner of LRP16. The interaction between these two proteins may establish a foundation for further studying the function of LRP16 0n embryonic development and cancer.
