可疑翼手参皂苷Colochiroside A体外抑制多种瘤细胞株增殖的研究

         

摘要

Objective :To study the antitumor activities in vitro of the triterpene glycoside Colochiroside A ( CA) from the sea cucumber Colochirus anceps.Methods : Detecting the deformation of mycelia germinated from.Conidia of Pyricularia oryzae and the antineoplastic activities to 5 types tumor cell ( BEL - 7402, HO - 8910, MDA - MB 435, MDA - MB -468.A431) and 2 types endothelial cell ( HMEC, HUVEC) in vitro was tested in demonstrating the effect of the saponin CA.Results : CA exhibited significant inhibiting activity against Pyricularia oryzae, the preliminary cytotoxic assay of CA exhihited significant cytotoxic activity against 5 types cultured tumor cell lines with the mean of IC50 1.75μg/ml -3.66μg/ml, and the preliminary cytotoxic assay of CA exhibited significant cytotoxic activity against 2 types cultured tumor cell lines with the IC50 of 5.63μg/ml, 2.55μg/ml respectively.Conclusion:Saporun CA exhibited remarkable antineoplaatic activities in vitro.%目的:研究可疑翼手参体内三萜皂苷Colochiroside A(CA)的体外抗肿瘤活性.方法:用稻瘟霉分生孢子模型对CA进行生物活性初筛,分别用MTT、SRB法检测CA对5种体外培养肿瘤细胞(BEL-7402、HO-8910、MDA-MB-435、MDA-MB-468、A431)和2种体外培养人血管内皮细胞(HMEC、HUVEC)的增殖抑制活性.结果:CA对稻瘟霉显示较强的抑制活性,最小活性浓度(MMDC)<7.9μg/ml;对5种体外培养人肿瘤细胞显示显著抑制增殖活性,IC50为1.75μg/ml-3.66μg/ml;对2种体外培养人血管内皮细胞均显示显著抑制增殖活性,IC50值分别为2.55μg/ml和5.63μg/ml.结论:CA体外对多种肿瘤细胞具有抑制活性,有进一步研究开发的前景.
