RGC32与转化生长因子β1在肺腺癌细胞增殖过程中调控关系的研究

摘要

目的：探讨 RGC32与转化生长因子β1（TGF -β1）在肺腺癌中表达的相关性，明确二者之间调控关系及其对肺腺癌细胞增殖的影响。方法：Real - time PCR 检测52例肺腺癌及相应癌旁组织中 RGC32与 TGF-β1的表达；应用 TGF -β1受体特异性抑制剂 SB -431542处理 A549细胞，检测处理后细胞中 RGC32基因的表达，并用 MTT 法测定细胞增殖活力，双标流式细胞术检测细胞凋亡。结果：RGC32和 TGF -β1在肺腺癌组织中表达均呈现显著增加，且二者呈正相关；SB -431542处理后，RGC32蛋白表达显著下调，A549细胞的凋亡增加，增殖活力降低。结论：RGC32和 TGF -β1在肺腺癌中均呈现表达上调，二者呈正相关。RGC32是TGF -β1信号通路下游重要的调控基因，能够参与肺腺癌细胞增殖能力的调节。%Objective:To research the relationshiP between RGC32 and TGF - β1 in lung adenocarcinoma,and the regulation between them,and their influence to Proliferation of A549 cells. Methods:All 52 cases of lung adeno-carcinoma and Peri - cancerous tissues were selected to detect the exPression of RGC32 and TGF - β1 by Real - time PCR. RGC32 was treated by SB - 431542(sPecific inhibitor of TGF - β1 recePtor)in A549 cell,the exPression of RGC32 was detected by Real - time PCR,the Proliferation of the treated cells was detected with MTT,the aPoPtosis was detected by flow cytometry. Results:ComPared with Peri - cancerous tissues,the exPression of RGC32 and TGF -β1 Presented an obvious uP - regulation in lung adenocarcinoma;there was a Positive relation between them. After treated with SB - 431542,the exPression of RGC32 decreased,the aPoPtosis increased significantly,and the Prolifera-tion decreased significantly. Conclusion:The exPression of RGC32 and TGF - β1 gene Presente an obvious uP - regu-lation in lung adenocarcinoma,there is a Positive relation between them. RGC32 is an imPortant downstream gene of TGF - β1 signal Pathway,and can regulate the Proliferation of lung adenocarcinoma cells.
