ATO联合TRAIL对肺癌裸鼠移植瘤抑制作用及机制研究

         

摘要

目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合三氧化二砷(ATO)对裸鼠肺癌移植瘤的抑制作用及其可能机制。方法将裸鼠肺癌移植瘤模型随机分为阴性对照组、TRAIL组、ATO低剂量组(ATO1组)、ATO高剂量组(ATO2组)、TRAIL联合ATO组,称取瘤质量并计算抑瘤率,采用免疫组化法检测半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表达。结果与阴性对照组比较,ATO1组、ATO2组对裸鼠肺癌移植瘤均具有抑制作用,TRAIL联合ATO组具有协同增效作用,并且上调Caspase-3的表达,差异均有统计学意义(P<0.05)。结论 ATO能增强TRAIL对裸鼠肺癌移植瘤的抑制作用,其机制可能与促进Caspase-3表达有关。%Objective To approach the inhibitory action and proposed mechanism of TRAIL combined with ATO on transplanted human lung cancer in nude mice. Methods The nude mice bearing A549 lung cancer samples were randomly divid-ed into the negative control group,group TRAIL,group ATO1,group ATO2 and TRAIL+ATO group. Tumor tissues were weighted and the inhibitory rate was observed. Immunohistochemistry was applied to detect the expression of Caspase-3 after treatments. Re-sults Compared with the negative control group,it had a strong the obviously action inhibition on transplanted human lung cancer of nude mice in group ATO1 and group ATO2 while a synergistic effect in group TRAIL+ATO ,and increased the expression of Caspase-3,whose difference had statistical significance(P<0.05). Conclusion ATO may inhibit the growth of transplanted hu-man lung cancer in nude mice through strengthening TRAIL,whose possible mechanism is potentially related to the expression of Caspase-3.
