FSTL1及MMP-2在子痫前期孕妇胎盘组织及滋养细胞中的表达及相关性研究

     

摘要

目的:探讨卵泡抑素样蛋白1(FSTL1)及基质金属蛋白酶2(MMP-2)在子痫前期孕妇胎盘组织及滋养细胞中的表达及相关性。方法以2014年1月至2015年9月在重庆医科大学附属第一医院产科住院行剖宫产术的子痫前期孕妇35例和正常足月妊娠行择期剖宫产术孕妇40例为研究对象,采用蛋白质印迹法检测正常晚期妊娠孕妇和子痫前期患者胎盘组织中FSTL1和MMP-2蛋白的表达;采用免疫荧光法检测正常培养和缺氧/复氧(H/R)处理的人早孕期绒毛膜外滋养细胞株HTR8/SVneo中FSTL1和MMP-2的表达;采用酶联免疫吸附试验(ELISA)检测两组孕妇血清FSTL1水平,分析FSTL1和MMP-2表达水平与子痫前期的相关性。结果与正常足月妊娠胎盘组织比较,子痫前期患者胎盘组织中FSTL1水平明显升高,而MMP-2水平明显下降,差异均有统计学意义(P<0.05)。与正常培养的细胞比较,H/R处理的细胞FSTL1蛋白水平明显升高,而MMP-2水平明显下降,差异均有统计学意义(P<0.05);FSTL1主要定位于HTR8/SV-neo细胞质中。正常足月妊娠孕妇血清FSTL1水平显著低于子痫前期孕妇,差异有统计学意义(P<0.01)。结论胎盘组织和血清中FSTL1和MMP-2水平的异常变化可能与子痫前期发病有关,二者可能存在协同作用。通过研究FSTL1和MMP-2的相互作用,可为子痫前期发病机制的研究提供新思路。%Objective To detect the expression and correlation of follistatin-like protein 1(FSTL1) and matrix metallo-proteinase-2(MMP-2) in placenta tissue and trophoblasts in preeclampsia pregnant women. Methods Thirty-five preeclampsia pregnant women and 40 normal full term pregnant women undergoing cesarean section in the obstetric department of the First Af-filiated Hospital of Chongqing Medical University from January 2014 to September 2015 were taken as the research subjects. Western blotting was adopted to detect the expression of FSTL1 and MMP-2 protein in placental tissues;the expressions of FSTL1 and MMP-2 in human early pregnancy extrachorionic trophoblastic cell lines HTR8/SVneo by normal culture and hypoxia/re-oxy-genation(H/R)treatment were detected by adopting immunofluorescence assay;the enzyme-linked immunosorbent assay(ELISA) was used to examine the levels of FSTL1 in serum in the two groups. The correlation between FSTL1 and MMP-2 expression levels with preeclampsia was analyzed. Results The expression level of FSTL1 in placenta tissues of preeclampsia patients was signifi-cantly increased compared with placenta tissue in full term pregnancy ,while the expression level of MMP-2 was significantly re-duced,and the differences were statistically significant(P<0.05). The expression level of FSTL1 protein in the cells of H/R treatment was significantly increased compared with that in the normal culture group ,while the expression level of MMP-2 was significantly reduced,and the differences were statistically significant(P<0.05);the positioning of FSTL1 was primarily located in cytoplasm of HTR8/SVneo. The serum FSTL1 level in the full term pregnant women group was significantly lower than that in the preeclampsia pregnant women,the difference was statistically significant(P<0.01). Conclusion The abnormal changes of FSTL1 and MMP-2 expression in placental tissues and serum may contribute to the pathogenesis of preeclampsia ,the two may have synergistic effect. Studying the interaction of FSTL1 and MMP-2 may provide a new idea for the research of preeclampsia pathogenesis.
