电针结合天麻素对局灶性脑缺血大鼠海马CA3区和表达的影响∗

     

摘要

目的::验证电针结合天麻素对局灶性脑缺血大鼠海马 CA3区生长相关蛋白-43(Growth-associated protein, GAP-43)和突触素(Synaptophysin,SYN)表达是否有协同增效作用。方法:SD 大鼠40只随机分为假手术组、模型组、电针组、天麻素组和电针结合天麻素组。采用线栓法阻塞大鼠右侧大脑中动脉复制局灶性脑缺血模型。造模成功后,按分组分别及同时给予电针刺激左侧曲池与合谷穴30min,天麻素注射液10mg/kg 腹腔注射,1次/d,连续治疗14d。免疫组织化学 SABC 法检测海马 GAP-43和 SYN 的表达。结果:与假手术组比较,模型组大鼠缺血侧海马 CA3区 GAP-43阳性神经元数量增多(P <0.05),SYN 阳性神经元数量减少(P <0.05);与模型组比较,电针组、天麻素组和电针结合天麻素组大鼠缺血侧海马 CA3区 GAP-43和 SYN 阳性神经元数量均显著增多(P <0.05);电针结合天麻素组 GAP-43和 SYN 阳性神经元数量均较电针组、天麻素组增多(均 P <0.05)。结论:电针结合天麻素对缺血侧海马 CA3区 GAP-43和 SYN 的表达具有协同增效作用,可更好地促进缺血损伤神经元的再生修复。%Objective:To validate whether there is synergy effect of electroacupuncture (EA)combined with gastrodin on the expression of (Growth-associated protein,GAP-43)and (Synaptophysin,SYN)in hippocampal CA3 in focal cerebral ischemia (FCI)rats.Methods:Forty SD rats were randomly divided into sham operation,model,EA,gastrodin and EA+ gastrodin groups.FCI rats were induced by middle cerebral artery occlusion with thread embolus.EA was applied to left side“Quchi”(LI 1 1)and“Hegu”(LI 4)after MCAO,gastrodin at 10mg/kg was given to rats by intrap-eritoneal.The expressions of GAP-43and SYN in hippocampal CA3 detected by immunohistochemical.Results:Com-pared with sham operation group,the number of GAP-43 positive neurons increased (P <0.05),the number of SYN positive neurons decreased (P <0.05).Compared with model group,the number of GAP-43 and SYN positive neurons increased (P <0.05 ),expression levels in the EA+gastrodin group increased significantly than the EA or gastrodin group (P <0.05 ).Conclusion:EA combined with gastrodin have synergistic effect on the expression of GAP-43 and SYN in the ischemic hippocampus CA3,can promote the regeneration and repair of injured neurons.
