Galectin-9对CD4+ T细胞亚群Th1/Th2/Th17/Treg免疫平衡的调节作用

Objective To study the immune regulation of Galectin-9 on the active CD4 + T subsets Th1/Th2/Th17/Treg and to explore the possible mechanisms. Methods Lymphocytes were harvested from wild-type C57BL/6 mice, from which CD4 + naiT cells were isolated with MACS and then stimulated to Thl/Th2/Thl7/Treg differentiation for 5 days. All subset cells were divided into 3 groups; Control group, Galectin-9 group and Galectin-9 +α-lactose group. The proportion of the subset cells was detected by FAC. Real time-PCR was used to detect the gene expression of transcription factors of the subset cells in each group. Results In the Galectin-9 group relative to the control group and Galectin-9 + α-lactose group, the proportions of Th1 and Thl7 cells were decreased, there was no significant change in Th2 cells and the expression of Treg cells was increased. The gene levels of T-bet and RORγt were deceased, no significant change in GATA-3 level was found and the expression of Foxp3 was increased. Conclusion Galectin-9 may inhibit the immune response of Th1 and Th17 cells and it has no effects on Th2 cells and can up regulate the immune response of Treg cells.%目的 研究Galectin-9对CD4+ T细胞亚群Th1/Th2/Th17/Treg免疫平衡的调节作用,并进一步探讨可能的作用机制.方法 获取野生型C57BL/6小鼠淋巴细胞,利用MACS分选CD4+ naT细胞,并分别向Th1/Th2/Th17/Treg进行定向分化,培养5 d.所有亚群细胞各分为3组:正常对照组、Galectin-9组和Galectin-9 + α-乳糖组.利用流式检测各组亚群细胞的比例,real time-PCR检测亚群细胞各组转录因子的基因表达水平.结果 与正常对照组和Galectin-9 + α-乳糖组相比,Galectin-9组Th1细胞和Th17细胞比例下降,Th2无明显变化,Treg细胞表达增加;T-bet和RORγt的基因表达水平均下降,GATA-3无明显变化,Foxp3的基因水平则升高.结论 Galectin-9能抑制Th1和Th17细胞的免疫应答反应,对Th2细胞无明显作用,同时能上调Treg细胞免疫应答.
