正交试验法优选泽兰迷迭香酸超声提取工艺

             

摘要

目的:通过正交试验法,优选泽兰(Lycopus Lucidus Turcz)中迷迭香酸的超声提取工艺。方法建立 HPLC-DAD 法测定泽兰中迷迭香酸含量的分析方法。以提取物中迷迭香酸的含量作为考察指标。采用单因素试验和正交试验考察提取溶剂、浓度、料液比、功率及提取时间对提取效率的影响,确定最佳提取工艺,并进行验证试验。结果最佳提取条件为:50%乙醇,功率198 W,料液比1∶15,提取时间30 min。验证试验表明此工艺条件下,泽兰中迷迭香酸的提取率为0.41%(RSD =0.11%,n =3)。结论优选后的提取工艺提取效率高,稳定性好,对今后提取、分离泽兰中迷迭香酸奠定基础。%Objective To optimize the extraction of rosmarinic acid from Lycopus Lucidus Turcz using the orthogonal test.Methods A method of HPLC-DAD was developed for determination of rosmarinic acid in L. Lucidus Turcz.The content of rosmarinic acid in the extract was used as the index of investigation.Then,the single factor test and orthogonal test were applied to investigate the influence of solvent,concentration,ratio of material to liquid,power and extraction time on the extraction progress,and the optimal extraction process was determined.Results Optimal extraction conditions were as follows:ethanol concentration 50%,micro-wave power 1 98 W,ratio of material to liquid (1 ∶1 5),extracting for 30 min.Validation test showed that aver-age extraction rate of rosmarinic acid was 0.41 % (RSD =0.1 1 %,n =3).Conclusion The optimized extrac-tion had high extraction efficiency and good stability which is useful for extraction and separation of rosma-rinic acid in L.Lucidus Turcz.
