白藜芦醇对人卵巢癌SKOV-3细胞形态及增殖的抑制作用

         

摘要

目的探讨白藜芦醇对人卵巢癌SKOV-3细胞形态学的影响及其增殖抑制作用。方法选取处于对数生长期的卵巢癌细胞株SKOV-3分为空白对照组和不同剂量白藜芦醇组。采用MTT法观察白藜芦醇对SK-OV-3细胞的抑制作用,计算出半数抑制浓度( IC50),依据IC50确定白藜芦醇的有效浓度;在光学显微镜下观察细胞形态学变化;Hoechst 33258染色后,荧光显微镜观察细胞核形态。结果与对照组比较,不同浓度白藜芦醇组处理后的人卵巢癌细胞SKOV-3的增殖明显受到抑制,并具有时间、剂量依赖关系;Hoechst 33258染色显示,不同浓度白藜芦醇培养SKOV-3细胞24 h后,荧光显微镜下均可观测到有不同程度的核皱缩及凋亡小体等强荧光团。结论白藜芦醇对人卵巢癌SKOV-3细胞形态学的影响明显,并可抑制其增殖。%Objective To investigate the effect of resveratrol on the morphology of human ovarian cancer SKOV-3 cells and its proliferation. Methods The ovarian cancer cell line SKOV-3 in logarithmic growth phase were selected and divided into control group and different dose group. MTT assay were used to observe the inhibitory effect of res-veratrol on SKOV-3 cells and calculate the half inhibitory concentration ( IC50 ) ,which determine the effective concen-trations of resveratrol. The morphological changes were observed under optical microscope;and nuclear morphology were observed by fluorescent microscope after Hoechst 33258 staining. Results Compared with the control group, the proliferation of ovarian cancer cell SKOV-3 was significantly inhibited by different concentration resveratrol group in time and dose-dependent manner. After co-culturing with different concentrations of resveratrol for 24 h,different degrees of nuclear shrinkage and apoptosis body stained with strong fluorophore were observed under the fluorescence microscope. Conclusion It was obviously that resveratrol could inhibit human ovarian cancer SKOV-3 cells’ prolifer-ation and have effect on its morphology.
