黄葵素对结核分枝杆菌的抑杀作用研究

         

摘要

Objective To evaluate the antituberculous activities of Huangkuisu (HKS). Methods ①Standard human strains of mycobacterium tuberculosis (H37RV) and multi-drug resistant TB strains were inoculated in different concentrations of HKS, blank control and positive TB drug culture medium to observe the growth of mycobacterium tuberculosis and evaluate the antibacterial activity in vitro. ② Certain number of standard strains of Mycobacterium tuberculosis H37Rv and drug-resistant strains were injected into the tail vein to establish a mouse model of mycobacterium tuberculosis infection. The mice were randomly divided into treatment group, rifampicin, HKS treatment group treated lasting for 6 weeks; the control group was given normal saline. The antibacterial activity in vivo were evaluated to compare the treatment results according to the pathological changes in lung and spleen tissues (HE staining and acid-fast staining) and charging amount of bacteria. Results ①the bactreial colonies were grew normally in model group, but were not grew inHKS and Rifampicin groups in cultured termination; resume training 40 d still no tuberculosis bacteria growth. HKS in 100 mg/mL was showing multi-drug-resistant strains of clinical suppression of better killing effect, compared with rifampin group, at each time point was indicative of statistical significance (P<0.05). ②in the mice infected by H37Rv, the lung and spleen tissue pathological changes in each treatment groups were light, the bacterial load in lung and spleen tissues were significantly reduced compared with the control group (P<0.05). Resistant strains of mice infected with mycobacterium tuberculosis in HKS group the bacterial load in lung and spleen tissues was significantly reduced compared with rifampin group and model control groups (P<0.05). Conclusion HKS is effective for killing and suppressing mycobacterium tuberculosis in vitro and in vivo, which canbe studied further as a drug against the drug fast tuberculosis.%目的 评价黄葵素(huangkuisu,HKS)抗结核分枝杆菌的活性.方法 ①采用人型结核分枝杆菌标准菌株(H37RV)、多药耐药结核菌株,分别接种于含有不同浓度黄葵素、空白对照和阳性药物的结核菌培养基,观察结核分枝杆菌的生长情况,评价其体外抗菌活性.②尾静脉注射一定数量的结核分枝杆菌标准株H37Rv和耐药株菌液,建立小鼠结核分枝杆菌感染模型.随机将模型小鼠分成利福平治疗组、黄葵素治疗组,持续治疗6周,对照组给予生理盐水治疗.以各组小鼠的肺组织和脾组织病理变化、组织荷菌量,比较各组治疗效果,评价其体内抗菌活性.结果 ①模型组菌落生长情况正常,黄葵素(100 mg/mL)、利福平(5 mg/mL)组结核分枝杆菌(H37RV)培养终止期未见菌落生长;恢复培养40d仍无结核菌生长.黄葵素在100 mg/mL时显示出对临床多耐药株的较好的抑杀作用,与利福平组比较,各时间点均显示出统计学意义(P<0.05).②结核杆菌H37Rv感染小鼠,各治疗组肺组织和脾组织的病理变化较轻,肺和脾组织荷菌量明显降低,与模型对照组比较有统计学意义(P<0.05).结核杆菌耐药株感染小鼠黄葵素治疗组小鼠组织器官的病理变化较轻,肺组织和脾组织的荷菌量明显减少,与利福平组、模型对照组比较有统计学意义(P<0.05).结论 黄葵素在体内外对结核杆菌有抑杀作用,可以作为治疗耐药株感染的候选药物进一步研究.
