表面活性剂降低烟碱对纤维素酶活性抑制作用的研究

     

摘要

为获得高效的烟秆酶解方法,缓解烟碱对于木质纤维素酶解体系的抑制作用,研究了烟碱对木质纤维素酶系中关键酶—纤维素酶的影响,在此基础上添加表面活性剂进一步分析纤维素酶活性的变化及酶解的效果.结果表明,0.1%底物质量分数的烟碱即会对纤维素酶活性产生显著的抑制作用,且随着烟碱质量分数的提高,抑制效果显著增强;非离子型表面活性剂PEG 6000、Tween 80和生物表面活性剂BSA、鼠李糖脂均能够不同程度降低烟碱对纤维素酶活性的抑制作用;Tween 80和BSA对烟秆酶解效果提升最显著,当添加量分别为0.03 g·g-1底物和0.001 g·g-1底物时,总糖质量浓度比对照分别提高了13.70%和10.34%,半纤维素转化率比对照分别提高了50.94%和40.04%,纤维素转化率没有明显变化.%In order to obtain the efficient method of tobacco enzymolysis this study examined the effects of reduing nicotine on cellulase activity inhibition by surfactant.Different surfactants including PEG 6000,Tween 80,BSA and Rhamnolipid were used to reduce nicotine production,subsequently allevi-ate the cellulase activity.The results showed that surfactants on the other hand,could alleviate the in-hibitory effect of nicotine on the activity of cellulase,0.1%nicotine could remarkably decrease the ac-tivity of cellulase activity.If the fraction of nicotine was increased,the inhibitory effects of nicotine on cellulase activity was significantly enhanced.Compare to control,when 0.001 g·g-1 of BSA and 0.03 g·g-1 of Tween 80 were added to the substrate,total reducing sugar concentration of tobacco stalk hydrolysate was increased by 13.70%and 10.34%,respectively.Compared to the control respectively.While hemicellulose conversion rate were increased by 50.94%and 40.04%with 0.03 g·g-1 of Tween 80 and 0.001 g·g-1 of BSA,respectively.But there was no obviously change in cellulose con-version rate.
