下调接头蛋白CrkII对喉癌Hep-2细胞生物学行为的影响

摘要

Objective:To investigate the effect of down-regulated junction protein CrkII on the proliferation, migration and invasion of laryngeal carcinoma Hep-2 cells.Methods:The expression of CrkII was reduced by RNA interference, and the plasmid was transfected into Hep-2 cells by liposome transfection.The levels of CrkII mRNA and protein were detected by real-time PCR and Western blot, respectively.MTT, Cell scratch and Transwell invasion assay were used to detect the changes of cell proliferation, migration and invasion ability after transfection.Results:Plasmid transfection could effectively inhibit the expression of CrkII mRNA and protein in Hep cells (P＜0.01).Proliferation, migration and invasion of Hep-2 cells decreased significantly (P＜0.01).Conclusion:CrkII gene silencing can significantly inhibit the proliferation, migration and invasion of Hep-2 cells.CrkII can promote the malignant biological behavior of Hep-2 cells in laryngeal carcinoma.%目的:探讨下调接头蛋白Crk II对喉癌Hep-2细胞增殖、迁移和侵袭能力的影响.方法:采用RNA干扰法敲低Crk II的表达, 以脂质体转染法将质粒导入Hep-2细胞中, 通过real-time PCR和Western blot分别检测各组细胞中Crk II mRNA和蛋白的水平, MTT、细胞划痕及Transwell侵袭实验检测转染后细胞增殖、迁移和侵袭能力的改变.结果:质粒转染后能有效的抑制Hep细胞Crk IImRNA和蛋白表达水平 (P＜0.01), Hep-2细胞增殖、迁移和侵袭能力明显降低 (P＜0.01) .结论:Crk II基因沉默可明显抑制Hep-2细胞的增殖、迁移与侵袭, 接头蛋白Crk II可促进喉癌Hep-2细胞的恶性生物学行为.
