牛蒡子苷对小鼠3T3-L1前脂肪细胞增殖与分化的影响及其作用机制

     

摘要

[目的]探讨牛蒡子苷( arctiin)对小鼠3T3-L1前脂肪细胞增殖、分化的影响及其作用机制.[方法]采用不同浓度牛蒡子苷作用于3T3-L1前脂肪细胞,四甲基偶氮唑盐(MTT)法检测其对细胞增殖的影响;油红O染色及分光光度法分析脂肪细胞的分化程度;实时荧光定量PCR检测细胞过氧化物酶增殖物激活受体γ(peroxisome proliferators activated receptorγ,PPARγ)和CCAAT/增强子结合蛋白α(CCTTA enhancer binging protein α,C/EBPα)的表达情况.[结果]牛蒡 子苷对3T3-L1细胞增殖无显著性影响(P>0.05),但可显著抑制3T3-L1前脂肪细胞向成熟脂肪细胞分化(P<0.05或P<0.01),且呈一定剂量依赖关系;与空白对照组比较,牛蒡子苷可显著降低PPARγ mRNA、C/EBPα mRNA表达水平(P<0.05或P<0.01).[结论]牛蒡子苷能抑制3T3-L1前脂肪细胞的成脂分化,其机制可能与其能抑制脂肪细胞分化相关调控因子PPARγ和C/EBPα的表达有关.%Objective To explore the effect and mechanism of arctiin on proliferation and differentiation of murine 3T3-L1 preadipocytes. Methods 3T3-L1 preadipocytes were cultured with arctiin in different dosages. Cell proliferation was analyzed by methyl thiazolyl tetrazolium colorimetric assay ( MTT). The degree of cell differentiation was evaluated by oil red 0 staining and spectrophotography. The expression of peroxisome proliferators activated receptor γ (PPAR-γ) and CCTTA enhancer binding protein α (C/EBPα) was quantitatively detected by real-time fluorescence quantitative-RT-PCR. Results Arctiin showed no significant influence on the proliferation of 3T3-L1 preadipocyles {P>0. 05}, but inhibited 3T3-L1 preadipocytes differentiating into mature adipocytes in a dose-dependent manner (P <0. 05 or P <0. 01). The mRNA levels of PPAR-y and C/EBPa were decreased by arctiin (P<0.05 or P <0.01 compared with the blank control group). Conclusion Arntiin can inhibit the differentiation of 3T3-L1 preadipocytes. The underlying mechanism may be related to inhibiting the expression of PPAR-y and C/EBPa which are associated with cell differentiation.
