柠檬酸滤纸面积及浓度对刺激健康人唾液分泌和唾液淀粉酶活性改变的影响

         

摘要

[目的]比较不同面积和浓度柠檬酸滤纸刺激对健康人唾液流率(mL/min)和唾液淀粉酶(salivary alpha-amylase,sAA)活性比值的影响.[方法]募集9例健康志愿者,分别给予0.2 mol/L 0.5 cm×0.5cm、0.2 mol/L 1 cm×1 cm、0.2mol/L 2 cm×2 cm、0.1 mol/L1 cm×1 cm和0.4mol/L1 cm×1 cm5种不同规格的柠檬酸浸泡滤纸刺激1 min,坐位采集刺激前2 min、刺激时1 min和刺激后2 min全唾液标本,并记录唾液量(mL),以碘-分光光度法测定sAA活性(单位),分别计算刺激时和刺激后的sAA活性比值以及刺激前、刺激时和刺激后唾液流率.[结果]对于不同面积0.2 rnol/L柠檬酸滤纸,以2 cm×2 cm刺激下唾液流率(刺激时:1.5833±0.813 9 mL/min;刺激后:0.675 0±0.258 0 mL/min)和sAA活性比值(刺激时:1.2967±0.3184;刺激后:1.4637±0.1093)增加最为显著(P<0.05);对于不同浓度1 cm×1 cm柠檬酸滤纸,以0.4 mol/L浓度刺激下唾液流率(刺激时:1.494 4±0.569 2 mL/min;刺激后:0.8528±0.404 7 mL/min)和sAA活性比值(刺激时:1.5571 ±0.458 5;刺激后:1.7246±0.4527)增加最为显著(P<0.05);对于刺激开始后不同采集时间段,除0.1 mol/L 1 cm×1 cm和0.2 mol/L0.5 cm×0.5 cm柠檬酸滤纸刺激时1min的sAA活性比值比刺激后2 min高之外,其他都表现为刺激后2 min的sAA活性比值比刺激时1 min显著增加(P<0.05);对于流率与sAA活性的关系,虽然刺激时1 min的唾液流率均高于刺激前2 min和刺激后2 min,但刺激后2 min的唾液流率与sAA活性正相关性(R2 =0.858 8),跟刺激前2 min(R2 =0.815 1)相近(P>0.05),且显著高于刺激时1min (R2=0.4625,P<0.05).[结论]健康人唾液流率和sAA活性与柠檬酸滤纸的面积和浓度密切相关;sAA活性比值实验唾液的采集,可以选择0.2 mol/L 2 cm×2 cm柠檬酸滤纸刺激舌尖1 min,以采集刺激后2 min的全唾液为宜.%Objective To compare the saliva flow rate and salivary alpha-amylase (sAA) activity ratio in healthy volunteers stimulated by citric acid filter paper at different concentrations and sizes. Methods Nine healthy volunteers in sitting position were stimulated for one minute by citric acid filter paper at different concentration and size combinations of 0. 2 mol/L - 0. 5 cm × 0. 5 cm, 0. 2 mol/L - 1 cm × 1 cm, 0. 2 mol/L - 2 cm × 2 cm, 0. 1 mol/ L - 1 cm × 1 cm and 0. 4 mol/L - 1 cm × 1 cm. The whole saliva sample of the volunteers was collected 2 min before stimulation, during stimulation for one minute and 2 min after stimulation. At the same time, the salivary volume was also recorded. Iodine spectrophotometry was adopted for the detection of sAA activity, and then the salivary flow rate and sAA activity ratio were calculated before stimulation, during stimulation and after stimulation. Results When the concentration of citric acid filter paper was set at 0. 2 mol/L, the paper size at 2 cm × 2 cm induced obvious increase of the saliva flow rate and sAA activity ratio (P <0. 05) , and the values were 1.583 3±0.8139 ml/min, 1. 296 7 ±0. 318 4 during the stimulation, and were 0. 675 0 ±0. 258 0 mL/min, 1. 463 7 ±0. 109 3 after stimulation. When the size of citric acid filter paper was set at 1 cm × 1 cm, the paper concentration at 0. 4 mol/L induced obvious increase of the saliva flow rate and sAA activity ratio (P < 0. 05) , and the values were 1. 494 4 ± 0. 569 2 mL/min, 1. 557 1 ± 0. 458 5 during the stimulation, and were 0. 852 8 ± 0. 404 7 mL/min, 1. 724 6 ± 0. 452 7 after stimulation. At different stimulation time, all types of citric acid filter paper except 0. 1 mol/L - 1 cm × 1 cm and 0. 2 mol/L -0.5 cm × 0. 5 cm had higher sAA activity ratio 2 min after stimulation than that during the stimulation ( P < 0. 05 ) .As for the relationship of salivary flow rate and sAA activity, though the flow rate during the stimulation was higher than that 2 min before stimulation and 2 min after stimulation, the flow rate was positively correlated with sAA activity 2 min after stimulation, and sAA activity was similar to that before stimulation ( P > 0. 05 ) and higher than that during the stimulation ( P < 0. 05 ) . Conclusion In healthy volunteers, the saliva flow rate and sAA activity are closely related with the size and concentration of citric acid filter paper. For the detection of sAA activity ratio, the saliva can be collected by putting 0. 2 mol/L - 2 cm × 2 cm of citric acid filter paper at the tongue tip 2 min after stimulating for one minute, and the whole saliva is the best.
