山葡萄‘双红’和‘双优’的试管快繁研究

         

摘要

The stems of Vitis amurensis two varieties ‘Shuanghong’and ‘Shuangyou’which have cold resistance were chosen as raw material.The influence of adventitious buds induction,proliferation and roo-ting culture,and the determination of the most appropriate matrix in transplanting stage were explored through several aspects including the choice of explants,sterilization time and the ratio of the different hor-mone.The results showed that the sterilization effect was best when the stems were disinfected with 75%alcohol first and then sterilized for 8 min in 0.1% HgCl2 solution.Survival rates were 86.67% and 90.00%for ‘Shuanghong’and‘Shuangyou’.The optimizing medium for proliferation of test tube plantlets was GS+NAA 0.1 mg/L,value multiples could be achieved 4.00.The best rooting medium was GS+IAA 0.05 mg/L,the root number could be over 9 per plant.The average length of adventitious roots was above 5 cm. The best matrix for transplanting plantlets was sand∶perlite∶vermiculite=1∶1∶2,survival rate up to 70.00%.%试验选用抗寒山葡萄‘双红’和‘双优’2个品种的茎段为外植体,分别从外植体的选择、消毒时间、不同激素配比等对不定芽诱导、增殖培养和生根培养效应等方面进行了研究,并确定驯化移栽的最适宜基质.结果表明:山葡萄‘双红’和‘双优’2个品种的外植体茎段,先用75%的酒精消毒30 s、再用0.1%的 HgCl2溶液消毒8 min获得的外植体消毒效果最佳,成活率分别为86.67%和90.00%;最适宜的增殖培养基为 GS+NAA 0.1 mg/L,增殖倍数可达到4.00;最适宜的生根培养基为GS+IAA 0.05 mg/L,可使单株平均根条数达9条以上,平均根长度达5 cm以上,最适宜的移栽基质为细沙∶珍珠岩∶蛭石=1∶1∶2,移栽试管苗成活率达70.00%以上.
