细胞支架中以丝蛋白为载体的生长因子负载方法研究

     

摘要

Using silk fibroin as a co-absorbed and protective carrier,a novel method of growth factor loading in tissue engineering scaffold was developed.The influence of the growth factor loading technology to the property of gelatin porous scaffold was studied.The porosities of gelatin scaffold and SF/EGF modified scaffold were over 90%.The water uptake increased 18 times after SF/EGF modification.Compressive tests showed an in-crease of compressive modulus at 26.1 kPa after SF/EGF modification.Elisa test displayed that active EGF could be detected in 4 weeks in vitro.The dose of released EGF from a milligram scaffold was in the range of 0.3-0.5 ng in every time intervals and the cumulative release was 3.473 ng during 28 d.MTT test and SEM ob-servation indicated that EGF loading scaffold improved the growth of epithelial cells.This growth factor loading method may be particularly useful in modification of tissue engineering scaffold.%在组织工程支架表面沉积含有生长因子的丝蛋白层,通过丝蛋白β构象转变,固定生长因子于支架表面.研究此种生长因子负载方法对明胶多孔支架性能的影响,结果表明,因子负载前后支架的孔隙率皆达到90%以上,吸水量皆为干燥时支架质量的18倍左右;力学测试表明,因子负载后支架的压缩模量增加至26.1 kPa;Elisa检测表明,在各取样时间点,每毫克支架缓释活性EGF的量约在0.3~0.5 ng之间,4周累积释放活性EGF达到3.473 ng;MTT实验与SEM观察表明,用此种方法负载因子于支架中,可以有效促进支架中细胞的生长.
