PD-L1在淋巴瘤中的表达及其对淋巴瘤增殖和化疗抵抗的初步研究

摘要

为了研究PD-L1(programmed death 1-1igad)对淋巴瘤增殖及对化疗药物的影响,探讨新的治疗方法,应用免疫组织化学方法和流式细胞术检测了PD-L1在淋巴瘤组织及细胞系中的表达情况,用流式细胞术分选的方法得到了高表达和低表达PD-L1的2组A20细胞株,将A20淋巴瘤细胞株接种到BALB/c小鼠身上构建荷瘤小鼠模型,观察肿瘤生长情况,最后检测了PD-L1对淋巴瘤常用化疗药顺铂的杀伤作用的影响.结果表明,PD-L1在淋巴瘤组织和细胞株中广泛高表达.应用流式细胞术筛选的PD-L1高表达细胞株在体内成瘤作用强于PD-L1低表达细胞株.结论:PD-L1可促进肿瘤的生长,同时通过抗凋亡抵制顺铂对肿瘤的杀伤作用.%In order to investigate the effect of PD-L1 on lymphoma cell proliferation and chemotherapy resistance, and explore a new and promising approach for therapy of lymphomas. The level of PD-L1 expression in tissues of patients with lymphoma and cell lines were detected by immunohistochemistry and flow cytometry. Both the stable high-PDL1-A20 cell strain and low-PDLl -A20 cell strain were sorted by flow cytometry, then inoculated into BALB/c mice as to establish a lymphoma bearing mouse model for observing tunor genesis. Finally, the effect of PD-L1 on routinely chemotherapy drug-cisplatin were observed. The results showed that the PD-L1 highly and widly expressed in lymphoma tissue and cell strains, the stable high-PD-Ll cell strain sorted by flow cytometry have higher lymphoma genesis than that of low-PD-Ll cell strain. It is concluded that PD-L1 can promote lymphoma genesis, meanwhile the PD-L1 may resist effect of cisplatin on lymphoma cells through anti-apoptosis.
