黄芪水蛭制剂对糖尿病肾病大鼠TGF-β1表达的实验研究

             

摘要

Objective: To investigate the effect of Huangqi shuizhi Preparation on expression of TGF-β1 of renal tissue in STZ-induced diabetic nephropathy rats, and further explore the mechanism of action of Huangqi shuizhi Preparation on the renal protection in STZ-induced Diabetic Nephropathy Rats. Methods: Fed with high fat diet and STZ produced by intraperitoneal injection in rats with diabetic nephropathy. Rats were randomly divided into six groups: normal control group,model control group,Chinese medicine group (Huangqi shuizhi Preparation group) .Leigongtengpian Duo DAI pian group,western medicine group (Lotensin group)and Chinese and Western medicine group (Huangqi shuizhi Preparation and Lotensin group). Rats were determined after treatment fasting blood glucose. Leave all rats's urine before the end of the experiment. And the 24 h urinary protein was determined. The expression of TGF-β1 of renal tissue in STZ-induced diabetic nephropathy rats was measured with immunohistochemical method. Results: After treatment for two months with renal sugar granule, the general situation in rats compared with the model rats were improved better; and the weight also gain significantly. Fasting blood glucose, 24h urinary protein in the treatment groups were significantly lower than the model group;and TGF-β1 of Renal Tissue in the model rats showed high expression, respectively significantly higher than every treatment groups. Conclusion: The possible mechanism of the renal protective effect of Leigongtengpian Duo DAI pian on STZ-induced Diabetic Nephropathy Rats is the inhibited expression of TGF-β1 in renal tissue in rats.%目的 研究黄芪水蛭制剂对糖尿病肾病大鼠TGF-β1表达的影响,探讨其在糖尿病肾病大鼠肾脏的保护机制.方法 采用高脂饲料喂养加STZ腹腔注射制作糖尿病肾病大鼠模型.分为正常对照组、模型对照组、中药治疗组(黄芪水蛭制剂)、雷公藤治疗组(雷公藤)、西药治疗组(洛汀新)、中西药治疗组(黄芪水蛭制剂加洛汀新),观察各组大鼠治疗后血糖变化,并于实验结束前进行留尿,测24h尿蛋白定量.免疫组化方法测定大鼠TGF-β1的表达.结果 经黄芪水蛭制剂治疗2个月后,各治疗组空腹血糖、24h尿蛋白定量均明显低于模型对照组.模型对照组TGF-β1表达显著高于各治疗组.结论 黄芪水蛭制剂对糖尿病肾病大鼠的肾保护作用可能与其抑制TGF-β1表达有关.
