针灸对1-氯-2,4-二硝基氯苯诱导的小鼠过敏性接触性皮炎的影响

     

摘要

Objective: To explore the effect of acupuncture and moxibustion on allerqic contact dermatitis (ACD) in mice and its potential molecular mechanism. Methods: The mice were divided into the normal control qroup,the model qroup,non Jinqluo acupuncture qroup,Jinqluo acupuncture qroup,Miconazole qroup,Jinqluo acupuncture plus Miconazole qroup,12 mice in each qroup. The levels of serum IL-2,IL-4,IL-7 and TNF-α were detected by enzyme linked immunosorbent assay (ELISA);the swellinq deqree of the skin of back and the orqan index of the mice were measured;the histopatholoqical chanqes of skin were detected by HE staininq;the expression levels of TLR4 and NF-kB in dermatitis tissue was detected by protein immunoblottinq(WB). Results:The results of HE staininq showed that there was no obvious chanqe in the skin structure in the normal control qroup;the skin tissue space of the model qroup was edema,and inflammatory cell qot infiltration;compared with the model qroup,the skin tissue of the non Jinqluo acupuncture qroup and Miconazole qroup did not chanqe siq-nificantly,the skin tissue edema and inflammatory cell infiltration in the Jinqluo acupuncture qroup and Jinqluo acupuncture plus Miconazole qroup were liqhten;the contents of serum IL-2,IL-4,IL-7 and TNF-α,the skin swellinq deqree,thymus index and spleen index,and the expression levels of TLR4 and NF-kB in the Jinqluo acupuncture qroup were siqnificantly lower than those in the model qroup and the non Jinqluo acupuncture qroup (P<0.05);the contents of serum IL-2,IL-4,IL-7 and TNF-α,the skin swellinq deqree,thymus index and spleen index,and the expression levels of TLR4 and NF-kB in the Jinqluo acupuncture plus Miconazole qroup were siq-nificantly lower than those in the model qroup and the Miconazole qroup (P<0.05);the contents of serum IL-2, IL-4,IL-7 and TNF-α,the skin swellinq deqree,thymus index and spleen index,and the expression levels of TLR4 and NF-kB in the Jinqluo acupuncture plus Miconazole qroup were not siqnificantly different from those in Jinqluo acupuncture qroup (P>0.05). Conclusion: Acupuncture and moxibustion have an improved effect on ACD mice,which may be achieved by requlatinq the activity of the TLR4/NF-kB siqnalinq pathway,providinq a certain basis for the clinical treatment of ACD.%目的 观察针灸对小鼠过敏性接触性皮炎(ACD)的影响,并探讨其潜在的分子作用机制.方法 将小鼠分为正常对照组、模型组、非经络针刺组、经络针灸组、咪康唑组、经络针灸+咪康唑组,每组12只. 采用酶联免疫吸附实验(ELISA)检测血清中白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-7(IL-7)、肿瘤坏死因子-α(TNF-α)的水平;检测各组小鼠背部皮肤肿胀程度以及脏器指数;HE染色检测皮肤组织病理学变化;蛋白免疫印迹(WB)检测皮炎组织中Toll样受体4(TLR4)、核因子kB(NF-kB)的表达水平. 结果 HE染色结果显示,正常对照组皮肤结构没有发生明显的变化;模型组皮肤组织间隙水肿,且炎性细胞浸润;与模型组相比,非经络针刺组和咪康唑组皮肤组织没有明显变化,经络针灸组和经络针灸+咪康唑组皮肤组织水肿和炎性细胞浸润情况减轻;经络针灸组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及皮肤中TLR4、NF-kB表达水平显著低于模型组与非经络针刺组(P<0.05);经络针灸+咪康唑组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及TLR4、NF-kB表达水平显著低于模型组和咪康唑组(P<0.05);经络针灸+咪康唑组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及TLR4、NF-kB表达水平与经络针灸组相比,差异无统计学意义(P>0.05). 结论 针灸对ACD小鼠具有改善作用, 这一作用可能是通过调控TLR4/NF-kB信号通路的活性实现的,为ACD的临床治疗提供一定的依据.
