N-乙酰半胱氨酸对大鼠肝纤维化肝中氧化及抗氧化的影响

         

摘要

[目的]探讨N-乙酰半胱氨酸(NAC)对大鼠肝纤维化肝中氧化及抗氧化体系的影响.[方法]用猪血清大鼠腹腔内注射复制免疫损伤性肝纤维化模型,以NAC为干预组,于第12周处死动物测定肝中过氧化氢酶(CAT)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、和维生素E(Vit E)、同时用Masson's三色染色胶原纤维半定量分析各组纤维化程度,并分析其相关性.[结果]NAC组肝中SOD、GSH、CAT和 VitE明显高于模型组,而MDA明显低于模型组.相关分析表明,各组纤维化程度与MDA呈非常显著的正相关(P<0.01),而与SOD、Vit E、GSH、CAT是显著负相关(P值均小于0.01).[结论]肝胶原量表达增加可能与脂质过氧化有关,NAC能有效降低肝内脂质过氧化,促使肝内氧化及抗氧化体系的恢复,减轻肝纤维化.%[Objective] To investigate the effect of N-acetylcysteine(NAC) on the oxidation and antioxidation system in the liver of rat hepatic fibrosis. [Methods] Immunolesion liver fibrosis models were copied by using pig serum injected into rat peritoneal cavity. NAC group was intervention group. Animals were killed at 12 weeks. Hydrogen peroxidase(CAT), malondialdehyde(MDA), superoxide dismutase(SOD), glutathione (GSH) and vitamine E(Vit E) in the liver were measured. Masson's dying was used to analyze the extent of hepatic fibrosis in each group, and their relationship was analyzed. [Results] SOD, GSH, CAT and Vit E in the liver of NAC group were significantly higher than those of model group, but MDA in NAC group was obviously lower than that in model group. The correlation analysis showed that the extent of liver fibrosis was significantly positively related with MDA( P<0.01), and negatively related with SOD, GSH, Vit E and CAT ( P <0.01, P <0.01, P <0.01, P <0.01, respectively). [Conclusion]The increase of hepatic collagen expression may be correlated with lipid peroxidation. NAC can effectively decrease lipid peroxidation, and restore the balance of oxidation and antioxidation system, and alleviate hepatic fibrosis.
