目的 检测RhoA和Ezrin蛋白各自在不同临床分期胃癌组织中的表达,探讨两者在胃癌发生和侵袭转移中的作用以及它们在其中的相互关系.方法 选取有完整临床资料的57例胃癌病例、23例非典型增生病例石蜡包埋组织,选27例非胃 癌组织病例的石蜡包埋组织标本作为对照组,应用免疫组织化学supervisionTM二步法分别检测3组病例中RhoA和Ezrin蛋白的表达情况.结果 Ezrin和RhoA蛋白在胃癌组织中表达率分别为86%和70.2%,在非典型增生组织中表达率分别为55.6%和51.9%,在 非癌胃组织中表达率均为8.7%.除胃癌和对照组中RhoA的表达差异无显著性意义外,3组病 例间的两种蛋白表达两两比较差异都有显著的统计学差异(P＜0.05).RhoA和Ezrin蛋白 表达与性别、年龄无关系,与细胞分化、病理分期、淋巴结转移有关系.胃癌组织中RhoA和 Ezrin蛋白表达呈正相关(Rs=0.509,P<0.01).结论 RhoA和Ezrin 蛋白表达均可能与胃癌的发生有关,均参与了胃癌的浸润转移及淋巴转移,可作为重要的生物学标志.%Objective To investigate the expression of Ezrin and RhoA proteins in gastric carcinoma tissue in different clinical stages, and anlyze their functions and relationships in the gastric carcinoma and develepment mechanism. The purpose was to study their roles in invasion and metastasis and to provide the new theoretical foundation. Methods The expressions of RhoA and Ezrin in 57 specimens of gastric carcinoma, 23 specimens of epithelial dysplasia and 25 specimens of normal gastric tissues were detected by immunohisto chemistry. Results ① The positive expression rates of Ezrin in normal gastric tissues, gastric epithelial dysplasia and gastric carcinoma were 8.7 ％,55.6％ and 86％, respectively, and those of RhoA were 8.7％, 51.9％ and 70.2％, respetively;② The expressions of Ezrin and RhoA were correlated to clinical stage, lymph node metastasisand differentiation ( P＜0.01 ); ③ The expression of Ezrin was positively correlated to that of Moesin in gastric carcinoma (P ＜ 0.01 ). Conclusions High expressions of Ezrin and RhoA are correlated with the clinical stage and lymph node metastasis in gastric carcinoma, and can be important rnarkers for forecasting metastasis and prognosis in gastric carcinoma.
