妊娠期甲状腺功能减退、亚临床甲状腺功能减退对仔鼠脑海马Bcl-2和Bax表达的影响

     

摘要

Objective To study the effects of pregency hypothyroidism and subclinical hypothyroidism on expression of Bcl-2 and Bax in offspring hippocampus.Methods SD female rats without thyroid by surgical resection were selected, and the rats were used to prepare the hypothyroidism model by subcutaneous injection of equal volume of normal saline, and made the subclinical hypothyroidism model by subcutaneous injection of L-T4, then, the models were mating with the normal male mice after 10 days, and then took the hippocampal cells of the offspring, and the normal pregnant mice offspring were included into the control group.Real-time PCR and Western Blot were used to detect the expressions of Bcl-2 and Bax gene and protein levels in hypothyroidism group, subclinical hypothyroidism group and control group at 14 days after birth.Results Compared with the control group, the expression of Bcl-2 was decreased in the hypothyroidism and subclinical hypothyroidism groups, and the expression of Bax was increased, and the differences were statistically significant.Compared with the subclinical hypothyroidism group and the control group, the changes of Bcl-2 and Bax expressions were more significant in the hypothyroidism group, and the differences were statistically.Conclusion At 14 days after the brith of pregnancy hypothyroidism and subclinical hypothyroidism offspring, the expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated, which provide clues for the study whether pregnancy hypothyroidism can induce apoptosis, as well as the mechaism.%目的 研究妊娠期甲状腺功能减退和亚临床甲状腺功能减退对仔鼠脑海马Bcl-2和Bax表达的影响.方法 采用手术完全切除甲状腺的SD雌鼠,皮下注入生理盐水制备甲状腺功能减退模型,皮下注射L-T4制备亚临床甲状腺功能减退模型,10 d后与正常雄鼠合笼交配,取仔鼠海马细胞,正常孕鼠后代仔鼠为对照组.采用Real-time PCR、Western Blot分别检测出生后14 d甲状腺功能减退组、亚临床甲状腺功能减退组与正常组仔鼠海马细胞Bcl-2和Bax基因水平和蛋白质水平表达的变化.结果 与正常对照组相比,甲状腺功能减退组及亚临床甲状腺功能减退组仔鼠Bcl-2基因水平和蛋白质水平下降,Bax基因水平和蛋白质水平升高,差异有统计学意义(P<0.05),且甲状腺功能减退组较亚临床甲状腺功能减退组和正常对照组仔鼠Bcl-2、Bax变化更明显,差异有统计学意义(P<0.05).结论 妊娠期甲状腺功能减退、亚临床甲状腺功能减退孕鼠的仔鼠出生后14 d时Bcl-2表达下调,Bax表达上调,上述结果为妊娠期甲状腺功能衰退孕鼠对后代脑发育过程中是否发生凋亡及其机制的研究提供线索.
