头穴透刺对脑出血大鼠脑组织中脾酪氨酸激酶表达的影响

     

摘要

Objective:To observe the expression of spleen tyrosine kinase (SYK) by needling GV20-to-GB7, and to explore its antagonism of nerve injury. Methods:156 male SD rats were randomly divided into the sham operation group, the model group, the scalp acupuncture group, and the inhibitor group. The cerebral hemorrhage model was established by autotransfusion. The inhibitor group was injected with Piceatannol (SYK inhibitor); the scalp acupuncture group was treated with needling GV20-to-GB7. Neurological function was tested after 6 h, 12 h, 24 h, and 72 h after modeling; the expression of SYK around brain hematoma was detected with Western Blot method and immunohistochemistry method. Results: Compared to the sham operation group, neurological function was significantly decreased and SYK expression was significantly increased in the model group at the same time point(P <0. 05). Compared to the model group, score of mNSS was obviously reduced, and SYK expression was obviously decreased in the scalp acupuncture group and the inhibitor group at 6 h, 12 h, 24 h, and 72 h after modeling; of which, there was no significant difference between the two groups (P >0. 05). Conclusion:SKY involved in the pathological process of the early cerebral hemorrhage, needling GV20-to-GB7 can reduce the SYK expression of the brain tissue in the rats with cerebral hemorrhage, thus to play the role in the antagonism of nerve injury.%目的:观察针刺"百会"透"曲鬓"穴对脑出血大鼠脑组织中脾酪氨酸激酶(SYK)表达的影响,以探讨针刺拮抗脑出血神经损伤的作用机制.方法:将雄性SD大鼠156只随机分为假手术组、模型组、抑制剂组和针刺组.采用大鼠自体血输注法建立脑出血模型,抑制剂组予以腹腔注射piceatannol(SYK抑制剂),针刺组选取"百会"透"曲鬓"穴进行针刺治疗.分别在造模后6、12、24、72 h时间点进行神经功能测定.分别应用免疫组化法和Western Blot测血肿周围脑组织SYK的表达水平.结果:与假手术组相比相同时间点模型组大鼠的神经功能明显下降,SYK水平明显升高(P<0. 05);与模型组相比相同时间点针刺组和抑制剂组m NSS评分明显减小,SYK水平显著降低(P<0. 05),并且抑制剂组与针刺组组间比较无显著性差异(P>0. 05).结论:SYK参与了早期脑出血的病理过程;针刺"百会"透"曲鬓"可能是通过降低脑出血大鼠脑组织中SYK表达水平,以拮抗脑出血神经功能损伤.
