研究了以醋酸纤维素/聚丙烯复合膜为载体的高碘酸钠氧化法固定化脂肪酶的固定化条件,采用响应面分析法对固定化条件进一步优化,并对固定化酶膜的酶学性质进行了讨论.结果表明,以固定化酶活力为指标,当高碘酸钠浓度为0.15 mol/L,活化为60 min时,浓度为0.013 g/mL的酶液与pH 8.0的磷酸盐缓冲溶液于戊二醛质量分数0.24%,温度4℃下,进行交联反应,反应3.1h后,获得的固定化酶活最高,为0.52 U/cm2.固定化酶膜的酶学性质为:固定化酶最适温度35℃,最适pH为9.0.%The lipase was immobilized on the acetylcellulose/polypropylene composite membrane by Sodium pe-riodate oxidation and the response surface analysis was used to further optimize immobilized conditions, and then discussed the properties of immobilized enzyme membrane. The lipase activities were 0. 52 U/cm2 ,and the optimal pH of the immobilized lipase was 9.0,the optimal operating temperature of the immobilized lipase was 35 ℃.
