一株疏水性解烃菌的分离鉴定及特性研究

         

摘要

A HH1 strain of hydrophobic alkane degrading bacteria HH1 was screened from petroleum-contaminated soils in Nanchong oil refinery. Through morphological, physiological and biochemical analysis,HH1 was catalasepositive, oxidase-negative, Gram-negative, peritrichous, with straight rods, no capsules, no endospores,0.5 - 0.8urn in diameter and 0.9 -2.0μm in length. And 1316bp of 16S rRNA sequences were amplified from HH1. By blast analysis, 16S rRNA showed 99.6% identity with Serratia proteamaculans wg-2 16S rRNA gene (accession no. EU627690) and Serratia proteamaculans 568 16S rRNA gene (accession no. CP000826). HH1 was finally identified as Serratia proteamaculans. This paper studied on the cell-surface hydrophobicity of HH1 by microorganism ad- hering to hydrocarbon (MATH) , the results showed that the cell-sufrace hydrophobicity of HH1 was 52.2% , and the cell-sufraee hydrophobieity changed with the variation of incubation time. The n-dodeeane degradation was 66. 1% after HH1 was growing on in hydrocarbon degradation medium with 1% (V/V) of n-docosane at 28℃ and 180r/min for 4d. It had potential in bioremediation of oil contaminated environment.%从南充市炼油厂附近被石油污染的土壤中分离获得一株疏水性烷烃降解菌HH1.通过形态学、生理生化特性研究发现菌株HHl为革兰氏阴性,中杆状,周生鞭毛,无荚膜,无芽孢,接触酶阳性,氧化酶阴性,菌体大小0.5—0.8μm×0.9—2.0μm;16SrRNA进化树分析发现HH1与Serratia proteamaculans wg-2和Serratia proteamaculans568的相似性分别为99.6%,菌株HHl归属为变形斑沙雷氏菌(Serratia proteamaculans).同时,采用微生物粘着碳烃化合物法(MATH)对菌株HHl表面疏水性进行研究,结果表明:菌株HHl的表面疏水性为52.2%,并随着培养时间增加,表面疏水性不断减小.在含有1%(V/V)正十二烷的无机盐培养基中接人1%种子液,于28℃、180r/min摇瓶振荡培养4d,正十二烷降解率可达66.1%.以上研究成果为石油污染的生物修复提供了一定的参考依据.
