目的 基于秀丽隐杆线虫生物模型,建立快速筛选中药活性的方法.方法 选用亨廷顿舞蹈症疾病模型AM141线虫株作为模型,以骆驼蓬多糖和小檗碱为代表药物,分别在固体及液体培养基中加入中药提取物或单体小分子化合物,以中药提取物溶解度、培养基有无污染、线虫polyQ荧光点数目、线虫形态及生长状态作为考察指标.结果 小檗碱20μM和骆驼蓬多糖10 mg/mL可显著抑制AM141线虫的polyQ聚集症状.液体培养基中的线虫易污染,长势不一,且中药提取物溶解性不好影响观察,所以筛选中药提取物时应使用固体培养基.结论 秀丽隐杆线虫模型用于药物筛选,快捷、方便、简单.%Objective Based on biological model of Caenorhabditis elegans,we establish a methodology which can rapidly screen activity of traditional Chinese medicine (TCM).Methods C. elegans strain AM141, a Huntington's disease animal model was applied for screening effective components, and polysaccharides of peganum harmala and berberine were used as representative drugs. Besides, herbs extracts or compounds were added into solid and liquid culture medium, respectively. Solubility of herbs extracts, culture media pollution free, number of polyglutamine fluorescence, morphology and development of worms were used as investigation index. Thus, establish rapidly screening methodology.Results Berberine (20μM) and polysaccharides of peganumharmala (10mg/ml) can significantly inhibit polyglutamine array aggregates phenotype. Worms in liquid culture medium were easy to be infected, resulting in different development state, as well as low solubility of the herbs extract, which affect observation, and difficult for TCM extract screening. Therefore, solid NGM culture medium was necessary. Conclusion With many advantages, including rapid, convenient,easy, Caenorhabditis eleganswas a useful model for drug screening.
