Objective To explore the protective effects of minocycline against apoptosis of hippocampal neurons and the relationships between doses. Methods Treated epileptic rats induced by pentylenetetrazol ( PTZ ) with different doses of minocycline using gavage method, the brain perfusion preparaed for paraffin sections and immunohistochemistry,the changes of apoptosis in rat hippocampus were observed by image analysis of HPIAS-1000 high-resolution color graphic analysis system pathology image analysis. Results Compared with PTZ group, apoptosis decreased in control group ( P < 0. 05 ); compared with PTZ group, apoptosis had no difference in MT1 group ( P > 0.05 ); compared with PTZ group, apoptosis decreased in MT2 group( P < 0. 05 ); compared with PTZ group, apoptosis increased in MT3 group ( P <0. 05 ). Comparison among the intervention group, compared with MT3 group and MT1 groups apoptosis decreased in MT2 apoptosis ( P<0.05 ). Conclusion Minocycline has neuroprotective effects in the appropriate concentration rangeo%目的 探讨米诺环素对神经元凋亡的保护作用及与剂量的关系.方法 采用戊四氮(PTZ)建立的癫(疒间)大鼠模型,用灌胃法给不同剂量的米诺环素后麻醉大鼠,灌注取脑进行石蜡切片制备及免疫组化检测,采用HPIAS-1000高清晰度彩色病理图文分析系统进行图像分析,观察大鼠海马区细胞凋亡变化.结果 对照组与PTZ组比较,对照组凋亡数目少(P<0.05);MT1组与PTZ组比较,细胞凋亡无差异(P﹥0.05);MT2组与PTZ组比较,MT2组细胞凋亡数少(P<0.05);MT3组与PTZ组比较,MT3组细胞凋亡多(P<0.05).各干预组之间比较,MT2组细胞凋亡较MT3组、MT1组都少(P<0.05).结论 米诺环素在适当的浓度范围内有神经元保护作用.
