With Thellungiella halophila as the material, GOD-POD method was adopted to determine the myrosinase activity in Thellungiella halophila rosette leaves and discuss the storage conditions of enzyme solution. As shown by the results, the myrosinase activity reached its maximum when substrate was catalyzed by I ml myrosinate system for 15 min, the myrosinate activity was significantly inhibited when storied in - 80 ℃. The improved method needed less plant sample and was easy to operate, which was applicable to the parallel determination of man y Thellungiella halophila samples.%采用盐芥为材料,利用葡萄糖氧化酶-过氧化物酶偶联体系(COD-POD)对莲座叶中提取的黑芥子酶活性进行了测定并对酶液存储条件进行了探讨.结果表明,在1 ml的酶液反应体系中与底物反应15 min时,黑芥子酶活性最高;酶液在-80℃保存活性显著下降.改进后的方法取样量小、操作较简便,适于较多盐芥样品的平行测定.
