基于ITS序列对秦岭4种未知真菌进行分子鉴定

     

摘要

[Objective] The aim was to provide the molecular basis for strain identification and specie division of macnvfiingi from Qinling mountain. [Method] 4 kinds of unknown fungus from Qinling mountain were taken aa the tested materials, their mycelium were separated and purified for genomic DNA extraction and ITS amplification of rDNA. The taxonomie siaius of the strains was determined based on ITS scquence. [Result] The results of identification showed that the 4 fungus were Cordyceps militant, Hericium erinaceum, Psathyrella candolleana and Trametes reap., and their aiaes were 541, 597, 682 and 602 bp reap. .[Conclusion]The study lay a foundation for development and utilization of macro-fungi from Qinling mountain.%[目的]为秦岭山区菌种鉴定和种类划分提供分子依据.[方法]以秦岭4种未知真菌(N1、N2、N3、N4)为试验材料,分离纯化苗丝体,提取基因组DNA并扩增其rDNA的ITS(Internal transcribed sequence,ITS)区,基于ITS序列对菌株的分类地位进行确定.[结果]经鉴定,N1、N2、N3、N4分别为北冬虫夏草(Cordyceps militaris)、猴头菇(Hericium erinaceum)、白黄小脆柄菇(Psathyrella candolleana)和云芝(Trametes),其大小分别为541、597、682和602 bp.[结论]该研究为秦岭山区大型真菌的开发利用奠定了基础.
