高效液相色谱法测定葡萄酒中11种有机酸含量

     

摘要

A rapid, sensitive and accurate solid phase extraction reverse phase high performance liquid chromatography-photodiode array detector( SPE-HPLC-PAD) method for simultaneous determination of eleven kinds of organic acids(oxalic acid, acetic acid, crylic acid, succinic acid, malonic acid, tartaric acid, fumaric acid, citric acid, propionic acid, malic acid and lactic acid) in four wines was studied. The analytes were separated by using a Hypersil C18 column(5 μm, 250 mm × 4.6 mm i. d. ) and methanol -0.5% Na2HPO4 as mobile phase with linear gradient elution. The eleven compounds were identified by retention time data and spectral characteristics and quantified by peak area. There were good linear correlations between the concentrations and peak areas of the eight components with the correlation coefficients of 0.996 2~0.999 5. The average recoveries were 85.5% -99.5% with relative standard deviations of 1.2% -4.3% (n = 5). The detection limits were 0.4-2.5 mg/L. The results also showed that it is a convenient, rapid, sensitive and accurate method. It can be used to analyze these organic acids in other complex matrices.%建立了固相萃取-高效液相色谱光电二极管阵列检测法测定葡萄酒中草酸、乙酸、丙烯酸、琥珀酸、丙二酸、酒石酸、富马酸、柠檬酸、丙酸、苹果酸和乳酸11种有机酸的方法,并用此方法测定了市售的4种葡萄酒中11种有机酸的含量.试验采用HypersilC18柱(5μm,250 mm×4.6 mmi.d.)分离,以甲醇-0.5%磷酸为流动相,线性梯度洗脱,以保留时间和特征光谱对分离出的组分予以定性确证,用峰面积进行定量.结果表明,11种有机酸组分的质量浓度与其峰面积在一定的范围内有良好的线性关系,相关系数为0.996 2 ~0.9995,11种组分的平均回收率为85.5% ~99.5%(相对标准偏差为1.2% ~4.3%),检测限为0.4~2.5 mg/L.该方法简便、快速、灵敏,也适用于其他复杂体系中草酸等11种有机酸的成分分析.
