[Objective] The aim was to optimize the fermentation conditions of cold-active catalase with Serratia liquefaciens CZA 1202 by response surface methodology. [ Method] The fermentation conditions of cold-active catalase with Serratia liquefaciens CZA 1202 were optimized by response surface methodology on the basis of the single factor test. [ Result ] The optimal fermentation conditions were that the fermentation temperature was 25℃; the initial pH was 8.0; the rotational speed was 190 r/min; the seed age was 36 h; the inoculum size was 10%. And under this condition,the enzyme activity of cold-active catalase was up to 288.96 U/ml. It was 10% higher than that before optimization,and basically according with the model predictive value of 290.15 U/ml. [Conclusion] The model built by this study was suitable for cold-active catalase production , and provided some theory basis for the large-scale production and application of cold-active catalase.%[目的]利用响应面法优化液化沙雷氏菌(Serratia liquefaciens)CZA 1202产低温过氧化氢酶的发酵条件.[方法]在发酵条件单因素试验的基础上通过响应面法优化液化沙雷氏菌CZA 1202产过氧化氢酶的发酵条件.[结果]最佳发酵条件为:温度25℃,pH 8.0,转速190 r/min,种龄36h,接种量10%,装液量100 ml(容量250 ml),在此条件下,最终低温过氧化氢酶酶活达288.96 U/ml,与优化前相比提高了10%,且与模型预测值290.15 U/ml基本吻合.[结论]该研究所建模型合适有效,为低温过氧化氢酶的规模化生产和应用提供了一定的理论基础.
