小鼠巨细胞病毒感染内耳的病理变化

         

摘要

目的 研究小鼠巨细胞病毒(MCMV)感染内耳的病理变化,并观察更昔洛韦(GCV)对MCMV感染所致内耳炎症反应的疗效.方法 72只小鼠随机均分为生理盐水(NS)10μl组(A组)、MCMV 10μl组(B组)、MCMV 10 IA+GCV 60 mg·kg-1·d-1(C组),每组均根据脑内、腹腔、耳内(仅左耳接种)注射部位不同进一步分组,每小组各8只.2周后耳蜗取材.结果 B组脑内、耳内感染方式下左耳耳蜗前庭膜增厚伴淋巴细胞浸润、蜗管顶部分泌表皮层纤维化改变;而B组腹腔、耳内感染方式下右耳及C组脑内、耳内感染方式下左耳耳蜗均未见上述病理变化.结论 脑内感染MCMV较腹腔、耳内(左耳)感染更易引起双耳前庭膜和蜗管的炎症反应.应用GCV干预对MCMV感染所引起的内耳炎症反应有一定的治疗作用.%Objective To investigate the pathological changes of the inner ear with murine cytomegalovirus(MCMV) infection and observe the therapeutic efficacy of ganciclovir (GCV) on MCMV-induced inflammatory reaction in the inner ear.Methods Seventy-two mice were randomly and equally divided into three groups of A(injected with NS 10 μl) ,B(injected with MCMV 10 μl) and C(injected with MCMV 10 μl+GCV 60 mg · kg-1 · d-1 ), which were all further randomly and equally divided into three subgroups according to different injection sites of brain,abdominal cavity and inner ear(only left ear), respectively.Two weeks later, the cochleas were taken out.Results The brain and ear infection in group B lead to vestibular membrane thickening, lymphocyte infiltration and fibrosis changes of cuticular layer in the cochear duct of left ear,which were not found in the right ear infected in the abdominal cavity and right ear of group B and the left ear infected in the brain and ear of group C.Conclusion MCMV infection in the brain more easily causes the inflammatory changes of vestibular membrane and cochear duct than that in abdominal cavity or inner ear.GCV treatment can decrease MCMV-induced inflammatory changes in the inner ear.
