吸水链霉菌ATCC29253产Hygrocins的条件优化

             

摘要

Streptomjces hygroscopicus ATCC29253 can product various antibiotics with strong bioactivities. To optimize the fermentation conditions of S. Hygroscopicus, incubation temperature, pH of medium and different aeration were tested. Meanwhile, the components of fermentation culture medium and different carbon and nitrogen sources were also detected through orthogonal test. The results showed that the optimized medium were as follows, 20.0 mlVL glycerol, 30.0 g/L pharmamedia, 1.0 g/L KH2PO4, 1.5 g/L K2HPO4, 5.0 g/L NaCl, 20.0 g/L mannitol, 5.0 g/L hydrolysis casein. The optimized medium highly improved the product ol hygrocins and rapamycin.%吸水链霉菌(Streptomyces hygroscopicus)ATCC29253能够产生多种具有较好生物活性的次级代谢产物,包括雷帕霉素、尼日利亚菌素、洋橄榄叶素、安莎类抗生素Hygrocins等.通过改变发酵的温度、pH及通氧量,探索菌丝体的最佳生长条件.利用正交试验优化改变培养基中各组分的含量,测试了不同的碳源和氮源对发酵的影响.优化后的培养基组成为甘油20.0 mL/L,棉子饼粉30.0 g/L,磷酸二氢钾1.0 g/L,磷酸氢二钾1.5 g/L,氯化钠5.0 g/L,甘露醇20.0 g/L,水解酪蛋白5.0 g/L.优化的发酵培养基不仅明显提高了hygrocins的产量,同时还较大幅度地促进了雷帕霉素的产生.
