鹿中结核分枝杆菌最佳DNA提取方法探索

摘要

对树脂法、改良CTAB法和国标法三种方法获得的鹿中结核分枝杆菌DNA进行比较,通过核酸扩增鉴定和琼脂糖凝胶电泳分析,发现改良CTAB法获得的基因组DNA最佳,配合PCR技术检测灵敏度达到100 fg,且能对低含量样本进行快速检测.%Three DNA extraction methods including resin method,improved CTAB method and national standard method were employed in obtaining DNA of mycobacterium tuberculosis in the deer respectively,followed the appraisal of amplification and agarose gel electrophoresis analysis.The results show that improved CTAB method can gain the highest result and quickly identify mycobacterium tuberculosis of the samples with low content along with the detection sensitivity of 100 fg.
