苦参碱预处理对大鼠脑缺血再灌注神经细胞凋亡及Bcl-2 Bax蛋白表达的影响

         

摘要

目的:观察苦参碱预处理后对脑缺血再灌注后脑神经细胞凋亡情况及凋亡相关蛋白表达的影响.方法:60只健康SD大鼠随机分为6组,分别为对照组、模型组、苦参碱组(4mg/kg,8mg/kg,16mg/kg),依达拉奉组(1mg/kg).采用改良线栓法构建脑缺血-再灌注损伤模型后,药物处理后,对各组大鼠不同时间段进行神经行为学评分;测定海马区脑组织脑神经细胞凋亡情况,和相关蛋白Bcl-2、Bax的表达情况.结果:不同剂量苦参碱各组大鼠神经行为学评分数值高于手术模型组(P<0.05),苦参碱各剂量组大鼠脑神经细胞凋亡率明显低于模型组(P<0.05),苦参碱各剂量组Bcl-2凋亡相关蛋白表达明显高于模型组(P<0.01),Bax凋亡蛋白表达明显低于模型组(P<0.05).结论:苦参碱可上调Bcl-2蛋白表达,下调Bax蛋白表达,调控神经细胞凋亡趋势,而产生神经保护的作用.%Objective:To observe the effects of matrine pretreatment on the apoptosis of nerve cells and the expression of Bcl-2 and Bax after cerebral ischemia and reperfusion in the brain. Methods:60 healthy SD rats were randomly divided into 6 groups, namely sham group, model group, matrine group ( 4, 8, 16 mg/kg), edaravone group (1 mg/kg). The cerebral ischemia reperfusion injury model was established by suture method in rats. Then neuro-behavior was scored in different time, brain cell apoptosis was determined and the expression of Bax and Bcl-2 were examined. Results:Compared by the model group, rats in matrine pretreat-ment group showed better neurobehavioral score ( P<0.05) , lower brain cell apoptosis rate ( P<0.05) , signifi-cantly higher expression of bcl-2 protein ( P<0.01) and lower expression of Bax protein ( P<0.05) . Conclu-sion:Matrine shows a neuro-protective role on cerebral ischemia and reperfusion injury in rats and it enhances the expression of Bcl-2 protein and inhibits the expression of Bax protein.
