大蒜素干预下大鼠血清对食管癌 EC109细胞凋亡及Survivin 表达的影响

             

摘要

目的:研究大蒜素干预下SD大鼠血清诱导食管癌EC109细胞凋亡及其对Survivin蛋白表达的影响。方法Ⅰ组:以普通灭活SD大鼠血清培养液培养食管癌EC109细胞;Ⅱ组:以大蒜素干预SD大鼠灭活血清培养液培养食管癌EC109细胞;采用AnnexinⅤ-EGFP/PI双染法和亚倍体峰法应用流式细胞仪检测2组食管癌细胞早期和中晚期凋亡率;Survivin蛋白的表达应用Westernblot方法检测。结果Ⅱ组细胞凋亡率明显高于Ⅰ组,差异有统计学意义( P <<0.05);Ⅱ组细胞中Survivin 蛋白的表达水平明显低于Ⅰ组,差异有统计学意义( P <0.05)。结论长期大蒜素干预下SD大鼠血清能促进食管癌EC109细胞的凋亡并抑制Survivin蛋白的表达。%Objective To investigate the effect of SD rats serum after intervention by allicin on the apoptosis of esophagus cancer EC-109 cells cell and the expression levels of Survivin.Methods The groupⅠ:esophagus cancer EC109 cells were cultured in normal inactivated SD rat serum medium;groupⅡ: esophagus cancer EC109 cells were cultured in inactivated SD rat serum medium plus allicin, then the cell apoptosis rates were detected by AnnexinⅤ-EGFP/PI double-standing and hypoploid peak method with flow cytometer, furthermore the expression levels of Survivin were measured by Western Blot.Results The cell apoptosis rates in group Ⅱwere significantly higher than those in groupⅠgroup ( P <0.05),however, the expression levels of Survivin protein in groupⅡwere significantly lower than those in groupⅠ( P 0.05).Conclusion The serum of SD rat after long-term intervention by allicin can promote the apoptosis of esophagus canc-er EC109 cells and can inhibit the expression of Survivin protein.
