食醋干预下大鼠血清对食管癌EC109细胞凋亡及Survivin表达的影响

         

摘要

目的研究长期高醋饮食 SD大鼠血清诱导食管癌 EC109细胞凋亡及其对 Survivin蛋白表达的影响。  方法采用普通 SD大鼠血清和长期高醋饮食条件下 SD大鼠血清培养食管癌 EC109细胞。I组:以含20%灭活普通 SD大鼠血清培养液培养食管癌 EC109细胞;II组:以含20%灭活长期高醋饮食干预条件下 SD大鼠血清培养液培养食管癌 EC109细胞;应用流式细胞仪分别采用 Annexin V-EGFP/PI双染法和亚倍体峰法检测两组食管癌细胞早期和中晚期凋亡率;应用 Western blot方法检测 Survivin蛋白的表达水平。  结果 II组细胞凋亡率明显高于 I组,有统计学差异(P <0.05);II组细胞中 Survivin蛋白的表达水平明显低于 I组,差异有统计学显著意义(P <0.05)。  结论长期高醋饮食 SD大鼠血清能促进食管癌 EC109细胞的凋亡并抑制 Survivin蛋白的表达。%Objective To study the effect of the SD rat' serum after the long-term high vinegar diet intervention on apoptosis and survivin protein expression in esophagus cancer EC109 cells. Methods Common SD rat' serum and the serum under the condition of long-term high vinegar diet intervention were used to cultivate esophagus cancer EC109 cells, with group I containing normal inactivated serum in culture medium, and group II containing the serum after the long-term high vinegar diet intervention. Annexin V-EGFP/PI double-staining and hypoploid peak method were used to detect the apoptosis rate using the flow cytometry. Western blot method was used to detect the Survivin protein expression. Results Cellular apoptosis rate of group II was significantly higher than that in group I (P< 0.05). The Survivin protein level of group II was lower than that of group I (P< 0.05). Conclusion The SD rat'serum under the condition of long-term high vinegar diet intervention can promote apoptosis and inhibit the Survivin protein expression in esophagus cancer EC109 cells.
