尾状核区注射法建立Wistar大鼠C6胶质瘤模型的实验研究

         

摘要

目的 探讨尾状核区注射法建立Wistar大鼠C6胶质瘤模型的技术要点及成瘤效果.方法 立体定向辅助下将C6细胞接种于Wistar大鼠右侧尾状核区,建立C6细胞胶质瘤鼠模型.共建立15只模型鼠,分别于接种的第7、14、21天行头颅MRI检查,3周后分批灌杀动物行病理HE切片观察、S100及GFAP免疫组织化学检测,了解肿瘤的生长特性和病理特征.对照组5只Wistar大鼠接种同体积培养液,同样方法进行检测.结果 用本法建立Wistar大鼠C6胶质瘤模型15只,成功率为100%.磁共振扫描显示接种第2周时10只大鼠有肿瘤生长,MR表现为T2高信号;第3周时全部有肿瘤生长,瘤周水肿明显.病理、免疫组织化学显示大鼠C6胶质瘤生长特性和病理特征与人脑胶质瘤相似.结论 该方法建立的胶质瘤模型稳定是进行胶质瘤研究的较好模型.%Objective To establish a rat model bearing brain glioma, and to investigate its experimental ap plication. Methods C6 cells were implanted into the unilateral brain hemisphere of 15 Wistar rats. MRI examination was performed on day 7, 14, 21. Three weeks after the implantation, the rates were killed for analysis of growth behav iors of the brain tumor and pathological features by HE, S100, GFAP. Results Fifteen Wistar rat model bearing brain glioma C6 cells were established, with the success rate of 100%. By day 14, the tumor became visible in 10/15 of the rats in at least two sections. By day 21, 15/15 of the rats showed space-occupying effect in the brain, and the tumor oc cupied over 1/2 of the hemisphere in 12/15 of the rats, and some tumor cells had migrated into the contralateral hemi sphere. Conclusion C6 glioma implantation model is easy, economical, practical and applicable for the glioma exper imental research.
