榆黄蘑子实体蛋白质提取工艺优化

         

摘要

To screen the best extract technology of total protein from P. citrinipileatus carpophore and lay the foundation for the development and utilization of its protein function, taking protein yield as index, protein was extracted from P. citrinipileatus carpophore by alkali method. On the basis of single experiment, including pH value, ratio of solid to liquid, ultrasonic time and ultrasonic power, an orthogonal experiment was carried out. It was concluded that the extraction rate of protein came to 19. 80 % on conditions as followed: ultrasonic power 90 W, the ratio of solid to liquid 1 : 25, extracting time 20 min, pH 14.%为筛选出榆黄蘑子实体中蛋白质提取的最佳工艺,为其蛋白质功能的开发利用奠定基础,以蛋白质得率为指标,采用碱提法提取榆黄蘑子实体中的蛋白质,在pH值、超声功率、提取时间、料液比等单因素试验的基础上进行正交试验,对蛋白质提取的工艺进行优化研究.结果表明:在超声功率90W、超声时间20 min、pH14、料液比1∶25条件下,榆黄蘑子实体中蛋白质得率可达19.80%.
