目的 探讨T细胞受体保守域Alpha 链基因(TCRα)-560C/T多态性与广西壮族人群IgA肾病的关系.方法 广西壮族IgA肾病患者(IgA肾病组)115例,广西壮族健康人(健康对照组)131例,采用PCR-RFLP法和PCR产物直接测序法对TCR-560C/T基因的单碱基突变多态性进行分析.结果 IgA肾病组中TCR-560C/T基因多态CT、CC、TT基因型频率分别为46.96%、23.48%和29.56%,其中C和T等位基因频率分别为46.96%和53.04%.健康对照组中,TCR-560C/T基因多态CT、CC、TT基因型频率分别为47.33%、25.19%和27.48%,其中C和T等位基因频率分别为48.85%和51.15%.IgA肾病患者组与健康对照组比较,基因型频率差异无统计学意义(P>0.05),等位基因频率之间差异无统计学意义(P>0.05).结论 TCT-560C/T等位基因可能不是广西壮族IgA肾病的易感基因.%Objective To study on the relationship between TCR-560C/T and polymorphism with susceptibility of IgA nephropathy of Zhuang nationality in Guangxi. Methods 115 cases with IgA nephropathy and 131 healthy controls of Guangxi Zhuang nationality were recruited. Genotypes of TCR-560C/T were analyzed by PCR-RFLP in 115 patients with IgA nephropathy and 131 controls. Results The frequencies of T TCR-560C/T gene genotypes CT,CC,TT in patients with IgA nephropathy were 46. 96% ,23. 48% ,29. 56% ,respectively. The frequencies of alleles C and T were 46. 96% and 53. 04% ,respectively. The frequencies of TCR-560C/T gene genotypes CT,CC, TT were 47. 33% ,25. 19% ,27. 48% in control group ,respectively. The frequencies of alleles C and T were 48. 85% and 51. 15% , respectively. The frequencies of TCR-560C/T genes genotypes in patients with IgA nephropathy were not significantly different from those in the controls( P >0. 05 ). The frequencies of alleles C and T were not significantly different( P >0. 05 )from those in the controls. Conclusion TCR-560C/T polymorphism may not association with IgA nephropathy of Zhuang nationality in Guangxi.
