Objective To study the metabolism method of( - )-epigallocatechin-3-O-gallate( EGCG )in vitro and its metabolites measured by high performance liquid chromatography( HPLC ). Methods EGCG was metabolized in rat liver homogenate in vitro with S-adenosylmethionine( SAM ), Mg2+ under appropriate temperature and pH values, and then the content of EGCG and its metaboltes were detected by HPLC. Results EGCG in vitro was metabolized efficiently. EGCG and its metabolites were analyzed by HPLC. Conclusion The metabolism method of EGCG in vitro is simple, convenient and accurate. It can be used to study the metabolism of EGCG effectively in vitro.%目的 研究体外代谢EGCG的方法,并用高效液相色谱法对EGCG及其产物进行测定.方法 在适当的pH和温度下,在正常大鼠的肝脏匀浆加入底物S-腺苷蛋氨酸(SAM)和Mg2+,进行EGCG体外代谢,用高效液相色谱法检测EGCG及其代谢物.结果 EGCG可在体外大鼠肝匀浆作用下进行有效代谢,可用高效液相色谱法对EGCG及其代谢产物进行检测.结论 这种体外代谢EGCG的方法简便灵敏,精密度高,可有效用于EGCG的体外代谢研究.
