保健食品中番茄红素测定方法的建立

         

摘要

High pertormance liquid chromatography (HPLC) method was developed for determination on lycopene in health foods.The samples were dissolved in gallic acid tert-butyl ester (TBG) dichloromethane solution and extracted by means of ultrasonic.The sample extract was cleaned up by polytetrafluoroethylene (PTFE) membrane.C18 Column (250 mm × 4.6 mam,5 μ.m) was as chromatographic separation column,and the mixed solution of acetonitrile and methanol was as mobile phase under elution.The detection on lycopene was carried out by HPLC.The external standard method was used for quantitative analysis.The calibration curves for lycopene were indicated in the range of 5.0 ~200 μg/mL,and the detection limit was 0.5 μg/mL.The recoveries of lycopene ranged from 81% to 96% with relative standard deviations 31% ~59%.The method was simple and rapid to meet the requirement for determination on lycopene in health foods.%对保健品中番茄红素的提取净化及液相色谱检测方法进行研究.样品用没食子酸叔丁酯(TBG)一一氯甲烷溶液溶解、超声波辅助提取、用PTFE滤膜过滤.采用C18色谱柱(250 mm×4.6mm,5um)分离,以乙腈和甲醇溶液洗脱测定,外标法定量.番茄红素在5.00 ~ 200μg/mL浓度范围的相关系数为0.993,检出限为0.5μg/mL,方法的回收率为81% ~96%,相对标准偏差为3.1% ~5.9%.该方法简便、准确,适用于保健品中番茄红素的检测要求.
