超临界萃取沙葱花挥发油的工艺优化及GC-MS分析

     

摘要

以野生沙葱花为原料,利用响应曲面法优化超临界CO 2提取沙葱花挥发油工艺,并对挥发油进行 GC—MS 检测分析。以萃取温度、萃取压力、萃取时间为影响因素,应用Box-Behnken 中心组合方法进行三因素三水平试验,以挥发油得率为响应值,进行响应面分析。沙葱花挥发油提取的最佳工艺条件为萃取温度39℃,萃取压力29 MPa,萃取时间1.2 h,流速20 L/h,挥发油得率预测值为19.36%,验证值为18.90%。通过 GC—MS 分析,分离出了29种组分,占总峰面积的96.67%,鉴定出了27种化合物,其中醛类化合物8种共占37.63%,芳香烃化合物共占30.52%,姜辣素占12.60%。%The volatile oil from the Allium mongolicum Regel flowers by supercritical-CO 2 fluid extraction (SFE ) was optimized using response surface methodology(RSM),and the volatile oil was i-dentified by GC—MS.The effects of the extraction pressure,extrac-tion temperature,extraction time on the yield of volatile oil were in-vestigated by a three-factor and three-level Box-Behnken central composite design with the extraction rate as the response value.The results showed that the optimum conditions for the extraction of Al-lium mongolicum Regel flower oil were as follows:extracting tem-perature 39 ℃,pressure 29 MPa,time 1 .2 h,CO 2 flux 20 L/h.Un-der such condition,the model-predicted and experimental values of oil yield were 1 9.36% and 18.90%.The 29 chemical components were separated by GC—MS,which were 96.67% of the total oil,and 27 compounds of them were also identified,including 8 aldehydes (37.63%),aromatic hydrocarbon compounds(30.52%)and Gingerol (12.60%).
