富锌酵母的筛选和培养条件优化

             

摘要

从实验室分离得到的16株酵母菌中,筛选出一株耐高浓度锌的酵母菌Y14,通过26S rDNA D1/D2区序列比对分析,确定该菌株为酿酒酵母(Saccharomyces cerevisiae).结果表明:培养基中添加300mg/L浓度的Zn,摇瓶培养48h时,Y14对Zn的吸附能力最高;正交实验结果显示,最适合Y14菌株吸附Zn的培养基组分为:葡萄糖20g/L、酵母粉10g/L、蛋白胨30g/L、KH2PO410g/L、Zn 300mg/L.优化后菌株对Zn吸附量提高50%左右,菌体锌含量达13.61mg/g.该菌株表现出对锌元素较强的吸附能力,具有进一步研究开发应用价值.%We screened a yeast strain which could tolerate high concentration of zinc from the 16 strains isolated in our laboratory. It was identified as saccharomyces cerevisiae by 26S rDNA D1/D2 sequence alignment. As the result showed: it got the maximum adsorption of Zn when Y14 strain was incubated in the medium which contained 300mg/L Zn at 30℃ for 48h. The components of medium for zinc adsorption were optimized by orthogonal test. The suitable medium components for zinc adsorption by Y14 strain was the follow(g/L): 20 of glucose, 10 of yeast ex-tract, 30 of peptone, 10 of KH2PO4 and added 300mg Zn. Zinc adsorption ability of the strain increased about 50% after optimization, and per gram cell contains 13.61mg of zinc. In conclusion, this strain showed strong ad-sorption ability of zinc, it has the significance for further research.
